温和灸对佐剂性炎症小鼠抗炎镇痛作用研究

摘要 目的明确艾灸对佐剂性炎症小鼠的抗炎镇痛作用,为进一步研究艾灸抗炎镇痛机制做铺垫。方法将基础足容积筛选合格的24只雄性C57BL/6J小鼠,随机分为对照组、模型组和模灸组,每组8只。模型组和模灸组小鼠右后足底注射20μL完全弗氏佐剂构建佐剂性炎症模型。造模后第4d开始干预,模灸组温和灸右侧“足三里”(ST36)。造模前、造模后以及开始干预后每5d测量各组小鼠足容积和痛阈;4周后取小鼠右侧膝关节滑膜,甲醛固定备检;SP法检测膝关节滑膜TNF-α的表达。结果①与对照组比较,模型组与模灸组造模后足容积显著增加(P<0.01);与模型组比较,模灸组在干预第19d足容积开始减小(P<0.05),在干预第24d至末次干预结束足容积显著减小(P<0.01)。②与模型组比较,模灸组自艾灸治疗第5d开始痛阈明显升高(P<0.01),治疗第19d时,模灸组小鼠痛阈与对照组无明显差异(P>0.05)。③治疗后模灸组小鼠关节滑膜中TNF-α表达较模型组低(P<0.01)。结论艾灸治疗具有明显的抗炎镇痛作用。 Objective To clarify the anti-inflammatory and analgesic effect of moxibustion on adjuvant-induced inflammation in mice,and to pave the way for further research on the anti-inflammatory and analgesic mechanism of moxibustion.Methods Twenty-four male C57BL/6J mice qualified for basal foot volume screening were randomly divided into control group,model group and model moxibustion group,with 8 mice in each group.The mice in the model group and the model moxibustion group were injected with 20μL of complete Freund's adjuvant in the right hind foot to build an adjuvant-induced inflammation model.The intervention began on the 4th day after modeling,and the moxibustion group in the model moxibustion group mildly moxibusted"Zusanli"on the right side(ST36).The foot volume and pain threshold of mice in each group were measured before and after model establishment and every 5 days after the intervention;after 4 weeks,the synovium of the right knee joint of the mice was taken and fixed with formaldehyde for examination;the SP method was used to detect the TNFαof the knee joint synovium expression.Results①Compared with the control group,the foot volume of the model group and the model moxibustion group increased significantly after modeling(P<0.01);From the 24th day to the end of the last intervention,the foot volume decreased significantly(P<0.01).②Compared with the model group,the pain threshold of the model moxibustion group was significantly increased from the 5th day of moxibustion treatment(P<0.01),and on the 19th day of treatment,there was no significant difference between the pain threshold of the model moxibustion group and the control group(P>0.05).③After treatment,the expression of TNF-αin the joint synovium of mice in the model moxibustion group was lower than that in the model group(P<0.01).Conclusion Moxibustion therapy has obvious anti-inflammatory and analgesic effects.
