circ_0000267通过靶向miR-1205调控胃癌细胞增殖、侵袭迁移的分子机制研究

摘要 目的:探讨circ_0000267是否通过靶向miR-1205调控胃癌细胞增殖、侵袭迁移。方法:选取三家医院(青海省西宁市中医院、中国人民解放军第九四一医院、青海省第五人民医院)2016年1月至2020年1月收治的30例胃癌患者的癌组织和癌旁组织;胃癌MKN-45细胞分为si-NC组、si-circ_0000267组、si-circ_0000267+anti-miR-NC组、si-circ_0000267+anti-miR-1205组。实时荧光定量PCR(RT-qPCR)检测circ_0000267和miR-1205的表达水平;MTT检测细胞增殖活性;Western blot法检测蛋白表达;Transwell检测细胞迁移和侵袭;双荧光素酶报告实验检测circ_0000267和miR-1205的靶向关系。结果:与癌旁组织比较,胃癌组织中circ_0000267表达水平升高,miR-1205表达水平降低(P<0.05)。抑制circ_0000267表达后,胃癌MKN-45细胞活性降低,CyclinD1表达水平降低,p21表达水平升高,迁移侵袭细胞数减少,MMP-2、MMP-9表达水平降低(P<0.05)。circ_0000267靶向调控miR-1205;下调miR-1205表达逆转了抑制circ_0000267表达对胃癌MKN-45细胞增殖和迁移侵袭的影响。结论:抑制circ_0000267可通过靶向调控miR-1205抑制胃癌细胞增殖、侵袭迁移。 Objective:To study whether circ_0000267 regulates the proliferation,invasion and migration of gastric cancer cells by targeting miR-1205.Methods:Thirty gastric cancer patients with cancer tissues and paracancerous tissues were selected from Xining Hospital of Traditional Chinese Medicine from January 2016 to January 2020;gastric cancer MKN-45 cells were divided into si-NC group,si-circ_0000267 group,si-circ_0000267+anti-miR-NC group,si-circ_0000267+anti-miR-1205 group.Real-time fluorescent quantitative PCR(RT-qPCR)detects the expression levels of circ_0000267 and miR-1205;MTT detects cell proliferation activity;Western blot detects protein expression;Transwell detects cell migration and invasion;dual luciferase reporter experiment detects circ_0000267 and miR-1205 targeting relationship.Results:Compared with adjacent tissues,the expression of circ_0000267 in gastric cancer tissues was increased,and the expression of miR-1205 was decreased(P<0.05).After inhibiting the expression of circ_0000267,the activity of gastric cancer MKN-45 cells was decreased,the expression of CyclinD1 was decreased,the expression of p21 was increased,the number of migration and invasion cells was decreased,and the expressions of MMP-2 and MMP-9 were decreased(P<0.05).circ_0000267 targets and regulates miR-1205;down-regulating the expression of miR-1205 reverses the effect of inhibiting the expression of circ_0000267 on the proliferation,migration and invasion of gastric cancer MKN-45 cells.Conclusions:Inhibition of circ_0000267 can inhibit the proliferation,invasion and migration of gastric cancer cells through targeted regulation of miR-1205.
