摘要
目的:探讨NLRP3-NF-κB炎症信号通路在糖尿病足(DF)伤口细胞焦亡中的作用。方法:纳入30例DF患者为观察组,以30例无糖尿病创伤患者为对照组,比较两组患者临床资料;采集伤口组织标本,行组织学观察,免疫荧光检测iNOS水平,ELISA检测IL-1β和IL-18蛋白水平的变化,PCR检测核因子κB-p65(NF-κB p65)、核因子κB抑制蛋白α(IκB-α)、凋亡相关斑点样蛋白(ASC)和半胱氨酸天冬氨酸蛋白酶1(Caspase-1)水平,Western blotting检测NLRP3表达水平。结果:对照组创面炎症反应较观察组平缓。观察组IL-1β和IL-18水平明显高于对照组(P<0.05)。观察组患者伤口NLRP3、NF-κB p65、IκB-α、ASC及Caspase-1水平明显高于对照组。结论:NLRP3-NF-κB炎症信号通路与DF伤口炎症反应密切相关,可能为DF溃疡的治疗提供新靶点。
Objective:To explore the The study of NLRP3-NF-κB inflammatory signal pathway of pyroptosis in diabetic foot ulcers.Methods:We enrolled 30 patients with diabetic foot ulcers and selected 30 trauma patients without diabetes as control group.The clinical data of the two groups was compared,and the granulated tissues were harvested and histologically analyzed in the both groups.And immune-histochemical and Enzyme-Linked Immunosorbent Assay(ELISA)was performed to evaluate the concentrations of IL-1βand IL-18.PCR was performed to evaluate the level of NF-κB p65,IκB-α,ASC and Caspase-1.Western blotting was performed to evaluate the level of NLRP3 inflammasome.Results:The inflammatory response in control group was milder than that in observation group.The level of IL-1βand IL-18 in the observation group was significantly higher than that in control group.Meanwhile the level of NLRP3,NF-κB p65,IκB-α,ASC and Caspase-1 from the wound tissue in the observation group was significantly higher than that in control group.Conclusion:NLRP3-NF-κB is closely related to the wound in diabetic foot and provides a new target to the therapy of diabetic foot ulcers.
