转移抑制基因KAI1负调控Wnt/β-catenin通路在抑制鼻咽癌转移机制中的研究

KAI1 negatively regulates the Wnt/β-catenin pathway in the mechanism of inhibiting the metastasis of nasopharyngeal carcinoma

目的:探讨转移抑制基因(KAI1)负调控Wnt/β-catenin通路抑制鼻咽癌转移的机制。方法:通过体外动物实验,构建人鼻咽癌细胞株裸鼠模型,并用注射高表达KAI1稳转细胞方法构建KAI1基因高表达模型,研究KAI1基因对该模型转移能力的影响。同时通过对细胞进行蛋白提取和蛋白质印迹法(Western Blot)检验,验证KAI1对Wnt/β-catenin通路的调控机制。结果:C组(KAI1实验组)肺转移病灶计数明显少于A组(空白组)和B组(内皮祖细胞组)。Western Blot检测结果显示C组Wnt-1和β-catenin相对表达量均低于A和B组。结论:KAI1基因可对Wnt/β-catenin通路起到负调控作用,为临床抑制肿瘤转移提供理论基础。

Objective:To explore the mechanism by which KAI1 gene negatively regulates Wnt/β-catenin pathway to inhibit the metastasis of nasopharyngeal carcinoma.Methods:In this experiment,through in vitro animal experiments,a nude mouse model bearing human nasopharyngeal carcinoma cell line was constructed,and a KAI1 gene high expression model was constructed by injection of high-expressing KAI1 stable transfected cells to study the effect of KAI1 gene on the metastatic ability of the model.At the same time,the cell protein extraction and Western Blot test were performed to verify the regulation mechanism of KAI1 on the Wnt/β-catenin pathway.Results:The count of lung metastases in the KAI1 experimental group was significantly less than that in the A control group(blank group)and B control group(endothelial progenitor cell group).And the detection of related protein expression by Western Blot showed that the relative expressions of Wnt-1 andβ-catenin in the same KAI1 experimental group were lower than those in the A and B groups.Conclusion:It is proved that KAI1 gene can negatively regulate the Wnt/β-catenin pathway.Provide a theoretical basis for clinical inhibition of tumor metastasis.
