摘要
【目的】为了揭示百合变温处理后相变的分子调控机制。【方法】以百合O×T杂种系中的杂交组合后代为试验材料,对百合仔球进行变温处理,通过qRT-PCR分析变温处理后仔球中miR 172与AP2-like基因家族的表达情况,并克隆了TOE3的同源基因LoTOE3。【结果】在变温处理条件培养下的百合仔球,15℃处理组的百合芽心分化较快,移栽后生长情况良好。qRT-PCR分析表明,LoTOE3的表达规律与miR 172的表达变化呈负相关,证明miR 172对百合LoTOE3基因的表达可能具有负调控作用。并成功克隆出LoTOE3基因的全长为1882 bp。【结论】为百合繁育的进一步研究,深入了解百合相变过程中的分子调控机制提供了理论参考及实践指导。
[Objectives]In order to reveal the molecular regulation mechanism of lilium lilium phase transition after temperature change treatment.[Methods]We used hybrid progenies from O×T hybrid lines as experimental materials to treat lily tuber at variable temperature.qRT-PCR was used to analyze the expression of miR 172 and AP2-like gene families in tuber at variable temperature,and cloned LoTOE3 homolog of TOE3.[Results]The buds of lilium tubers cultured at variable temperature showed rapid differentiation and good growth after transplanting at 15℃.qRT-PCR analysis showed that the expression pattern of LoTOE3 was negatively correlated with the expression of miR 172,which proved that miR 172 had a negative regulatory effect on the expression of the LoTOE3 gene in lily.The total length of LoTOE3 gene was 1882 bp.[Conclusion]This study provides theoretical reference and practical guidance for further research on lily breeding and further understanding of molecular regulation mechanisms during lily phase transition.
