扶正抗癌汤联合顺铂上调PTEN介导PI3K/AKT信号通路抑制卵巢癌细胞增殖

Fuzheng Kangai Decoction combined with cisplatin up-regulates PTEN-mediated PI3K/AKT signaling pathway to inhibit the proliferation of ovarian cancer cells

摘要 目的探究扶正抗癌汤联合顺铂对卵巢癌细胞增殖的影响,探索PETN介导的PI3K/AKT信号通路在该调节过程中发挥的作用。方法将32只裸鼠随机分为对照组、中药组、顺铂组、中药联合顺铂组,每组8只,每日测量瘤体体积,取材后,称量瘤体重量;免疫组织化学染色观察Ki67阳性表达情况;流式细胞技术检测CD4^(+)、CD8^(+),计算比值;ELISA法检测血清中TNF-α和IL-2水平;Western blot法检测瘤体中PTEN、PI3K、p-Akt和Akt蛋白的表达。结果扶正抗癌汤联合顺铂治疗抑制瘤体生长,降低肿瘤组织Ki67的表达,升高血清中TNF-α和IL-2水平,提高机体免疫功能,促进瘤体中PETN的表达、抑制PI3K、p-Akt的表达。结论扶正抗癌汤联合顺铂可以通过上调PTEN抑制PI3K/AKT信号通路来抑制卵巢癌细胞的增殖,抑制肿瘤生长。 Objective To explore the effect of Fuzheng Anticancer Decoction combined with cisplatin on the proliferation of ovarian cancer cells,and explore the role of PETN-mediated PI3K/AKT signaling pathway in this regulation process.Methods 32 nude mice were randomly divided into a control group,Fuzheng group,Cisplatin group and Combined group,with 8 in each group.The tumor volume was measured every day,and the weight of the tumor was measured after the material was taken.Immunohistochemical staining was used to observe the positive expression of Ki67.CD4^(+)and CD8^(+)were detected by flow cytometry,and the ratio was calculated.ELISA was used to detect the levels of TNF-αand IL-2 in serum.Western blot was used to detect the expression of PTEN,PI3K,p-Akt and Akt in tumors.Results Fuzheng Anticancer Decoction combined with cisplatin treatment inhibits tumor growth,reduces the expression of Ki67 in tumor tissues,increases the levels of TNF-αand IL-2 in serum,improves the body’s immune function,upregulates the expression of PETN in tumors,and inhibits the expression of PI3K and p-Akt.Conclusion Fuzheng Anticancer Decoction combined with cisplatin can inhibit the proliferation of ovarian cancer cells and inhibit tumor growth by up-regulating PTEN to inhibit PI3K/AKT signaling pathway.
