摘要
目的 探讨金丝桃苷对支原体肺炎模型小鼠TLR-4/MyD88/NF-κB信号通路的影响。方法60只BALB/c小鼠通过随机数表法分为对照组(Control)、模型组(MP)及金丝桃苷组(Hyp),每组20只,采用支原体菌液滴鼻感染建立支原体肺炎小鼠模型。HE染色观察各组小鼠肺组织病理学变化;全自动生化分析仪检测血清中C-反应蛋白(CRP)、丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、乳酸脱氢酶(LDH)和心肌型肌酸激酶同工酶(CK-MB)水平,检测肺组织中活性氧簇(ROS)、超氧化物歧化酶(SOD)和谷胱甘肽(GSH)水平;酶联免疫方法(ELISA)检测血清白细胞介素1β(IL-1β)、白细胞介素-6(IL-6)与肿瘤坏死因子-α(TNF-α)的含量;Western blot方法检测各组小鼠肺组织TLR-4、MyD88与NF-κB蛋白的表达。结果 金丝桃苷能够减轻MP感染小鼠肺组织的炎症反应;降低MP感染小鼠血清CRP、ALT、AST、LDH和CK-MB表达水平(P<0.01);降低血清IL-1β、IL-6和TNF-α含量(P<0.01);降低MP感染小鼠肺组织ROS表达水平,上调SOD和GSH表达水平(P<0.01);降低TLR-4、MyD88与NF-κB蛋白的表达(P<0.01)。结论 金丝桃苷能够减轻肺炎支原体感染小鼠的炎性反应,其作用机制可能与抑制TLR-4/MyD88/NF-κB信号通路有关。
Objective To investigate the effect of hyperin on TLR-4/MyD88/NF-κB signaling pathway in mycoplasma pneumonia model mice. Methods Sixty BALB/C mice were divided into control group(Control group), model group(MP) and hyperin group(Hyp) by random number table method, with 20 mice in each group. Mycoplasma pneumonia mouse model was established by mycoplasma droplets infection. HE staining was used to observe the pathological changes of lung tissues in each group. The levels of C-reactive protein(CRP), alanine aminotransferase(ALT), aspartate aminotransferase(AST), lactate dehydrogenase(LDH) and cardiac creatine kinase isoenzyme(CK-MB) in serum were detected by automatic biochemical analyzer. The levels of reactive oxygen species(ROS), superoxide dismutase(SOD) and glutathione(GSH) in lung tissue were detected. The contents of interleukin-1 β(IL-1β), interleukin-6(IL-6) and tumor necrosis factor(TNF-α) in serum were determined by enzyme-linked immunosorbent assay(ELISA). The expressions of TLR-4, MyD88 and NF-κB in lung tissues of mice in each group were detected by Western blot. Results Hyperin can reduce the inflammatory response in lung tissue of MP-infected mice. The levels of CRP, ALT, AST, LDH and CK-MB in serum of MP-infected mice were decreased(P<0.01). The levels of IL-1β, IL-6 and TNF-α in serum were decreased(P<0.01). The level of ROS was decreased, and the level of SOD and GSH were increased(P<0.01). The expressions of TLR-4, MyD88 and NF-κB were decreased(P<0.01). Conclusion Hyperin can reduce the inflammatory response of mycoplasma pneumoniae infected mice, and its mechanism may be related to the inhibition of TLR-4/MyD88/NF-κB signaling pathway.
