鱼腥草素钠通过抑制STAT3/NF-κB信号通路减轻哮喘幼鼠的肺内炎症反应 被引量:5

摘要 目的 探讨鱼腥草素钠对哮喘幼鼠肺内炎症反应的影响及可能机制。方法 45只SPF级BALB/c幼鼠随机分为对照组(Control)、哮喘组(Model)和鱼腥草素钠组(SH),每组15只,采用卵清蛋白致敏(OVA)制备幼鼠支气管哮喘模型。模型制作成功后,采用小动物肺功能分析仪检测小鼠气道高反应性;对各组小鼠支气管肺泡灌洗液(BALF)进行总细胞计数和白细胞分类计数;ELISA方法检测BALF中白介素-6(IL-6)和肿瘤坏死因子α(TNF-α)的含量;HE染色观察各组小鼠肺组织病理学变化;免疫组织荧光方法与Western blot方法检测各组小鼠肺组织STAT3与NF-κB p65蛋白的表达。结果 与哮喘组相比,鱼腥草素钠组幼鼠气道高反应明显降低(P<0.01);BALF中细胞总数及嗜酸性粒细胞数明显降低(P<0.01), IL-6与TNF-α的含量明显降低(P<0.01)。HE染色结果显示,鱼腥草素钠能够减轻哮喘幼鼠肺组织的炎症反应。免疫荧光化学方法与Western blot结果显示,鱼腥草素钠能够减轻哮喘幼鼠肺组织STAT3与NF-κB p65蛋白的表达(P<0.01)。结论 鱼腥草素钠能够通过抑STAT3/NF-κB信号通路抑制哮喘幼鼠的肺内炎性反应。 Objective To investigate the effect of sodium houttuynia on lung inflammation in asthmatic young mice and its possible mechanism. Methods Forty-five SPF BALB/C young mice were randomly divided into control group,asthma group and sodium houttuyfonate group(SH), with 15 young mice in each group. The young mice were sensitized by ovalbumin(OVA) to establish bronchial asthma model. After the model was made successfully, the airway hyperreactivity of mice was detected by small animal pulmonary function analyzer. Total cell count and leukocyte classification count of bronchoalveolar lavage fluid(BALF) were performed in each group. The contents of interleukin-6(IL-6) and tumor necrosis factor α(TNF-α) in BALF were determined by ELISA. HE staining was used to observe the pathological changes of lung tissues in each group. The expression of STAT3 and NF-κB P65 protein in lung tissues of each group was detected by immunofluorescence and Western blot. Results Compared with asthma group, airway hyperreaction of mice in SH group was significantly reduced(P<0.01). The total number of cells and eosinophils in BALF was decreased(P <0.01), and the contents of IL-6 and TNF-α in BALF was decreased(P<0.01). The results of HE staining showed that sodium houttuynitin could reduce the inflammatory response of lung tissue in asthmatic young mice. The results of immunofluorescence and Western blot showed that sodium houttuynia could reduce the expression of STAT3 and NF-κB P65 in lung tissues of asthmatic young mice(P<0.01). Conclusion Sodium houttuynia can inhibit the inflammatory response in lung of asthmatic young mice by inhibiting the STAT3/NF-κB signaling pathway.
