基于网络药理学的健脾清化方治疗2型糖尿病的作用机制研究

摘要 目的 探究健脾清化方治疗2型糖尿病的潜在活性成分和作用机制。方法 采用TCMSP数据库检索健脾清化方中党参、黄芪、山药、葛根、黄芩、黄连、黄精、鬼箭羽所含有的化学成分及作用靶点,Uniprot database用于对靶点基因的校正,Cytoscape software分析药材、成分及靶点网络,通过使用String database和Cytoscape software确立蛋白互作网络,利用David数据库对作用靶点进行GO和KEGG功能富集分析。结果 药材-成分-靶点网络含131个化合物节点和257个靶点节点,Degree值排名前5位的化合物分别槲皮素、β-谷甾醇、山奈酚、豆甾醇和刺芒柄花素。蛋白互作网络中治疗2型糖尿病靶点可能是TNF、IL-6、VEGF、AKT1、FOS、TP53、EGFR、EGF、MAPK14、MAPK1等。GO功能分析在生物功能方面涉及NO生物合成过程的正调控、药物反应、蛋白质磷酸化的正调控等;分子功能方面涉及相同蛋白和酶结合等;细胞组成方面涉及细胞外间隙、细胞外区、血小板α-膜粒膜蛋白等。KEGG通路富集发现上述靶点在肿瘤、美洲锥虫病、乙型肝炎等疾病方面发挥重要的调控作用。结论 健脾清化方可通过多成分、多靶点和多途径从而发挥对2型糖尿病的治疗作用。 Objective To explore the potential active ingredients and mechanism of Jianpi Qinghua Recipe in the treatment of type 2 diabetes.Methods The TCMSP database was used to search the chemical components and targets of Codonopsis,Astragalus,Chinese yam,Pueraria lobata,Scutellaria baicalensis,Rhizoma Coptidis,Rhizoma Polygonati and Guijianyu in Jianpi Qinghua Recipe.Uniprot database was used to correct target genes.Cytoscape software analyzes medicinal materials,components and target networks.By establishing protein interaction networks using String database and Cytoscape software,GO and KEGG functional enrichment analysis was performed on the targets using the David database.Results The medicinal material-component-target network contains 131 compound nodes and 257 target nodes.The top 5 compounds in Degree value were quercetin,beta-sitosterol,kaempferol,stigmasterol and formononetin.The targets for the treatment of type 2 diabetes in the protein interaction network may be TNF,IL-6,VEGF,AKT1,FOS,TP53,EGFR,EGF,MAPK14,MAPK1.GO functional analysis involves positive regulation of NO biosynthesis,drug response,and positive regulation of protein phosphorylation in terms of biological functions.Molecular function involves the same protein and enzyme binding.In terms of cell composition,it involves extracellular space,extracellular region,platelet α-granule membrane protein and so on.KEGG pathway enrichment found that the above targets play an important regulatory role in cancer,Chagas disease,hepatitis B and other diseases.Conclusion Jianpi Qinghua Recipe can play a role in the treatment of type 2 diabetes through multi-component,multi-target and multi-way.
