摘要
以苦瓜、枸杞子、桔梗、葛根、干姜、黄精为原料制备苦瓜本草酵素,并对苦瓜本草酵素的功能活性成分进行测定;通过构建2型糖尿病小鼠模型,评价苦瓜本草酵素辅助二甲双胍对2型糖尿病小鼠的降糖作用。将30只2型糖尿病小鼠随机分为2型糖尿病模型组(n=10)、二甲双胍组(n=10)、二甲双胍+酵素组(n=10),另设空白对照组(n=10)。受试小鼠在试验过程中均正常灌胃二甲双胍,处理组小鼠每日灌胃苦瓜本草酵素稀释液10 mL/kg·bw(含苦瓜本草酵素原液0.2mL),空白对照组和2型糖尿病模型组小鼠每日灌胃等量生理盐水。试验周期为30 d,定期记录小鼠的摄食饮水量及体重。试验后分别对各组小鼠进行血液指标检测。结果表明,在苦瓜本草酵素协同二甲双胍干预下,2型糖尿病小鼠饮水量、摄食量和血糖水平含量均明显降低,小鼠葡萄糖耐受力增强,血脂水平有所下降。苦瓜本草酵素的辅助降糖效果明显,为开发降糖产品奠定了理论基础。
Momordica charantia herbal enzyme was prepared with Momordica charantia,Lycium barbarum,Platycodon grandiflorum,Puerariae Radix,dried ginger and Polygonatum.The functional active components of Momordica charantia herbal enzyme were determined.Then,the type 2 diabetic mice were constructed to evaluate the hypoglycemic effect of bitter melon herb enzyme assisted metformin.T2D mice were randomly divided into T2D mice model group(n=10),metformin group(n=10),metformin+Momordica charantia herbal enzyme group(n=10),and blank control group(n=10).The T2D mice were given metformin enteric coated tablets during the experiment,and T2D mouse was fed with Momordica charantia herbal enzyme 10 mL/kg·bw(containing 0.2 mL of Momordica charantia herbal enzyme original solution)daily.The other control and model groups were given normal saline every day.The test cycle was 30 days.The water/food intake and body weight of mice were recorded regularly.After the test,the blood indexes of mice in each group were detected.The results showed that the water/food intake and blood glucose level of diabetes mice obvious decreased,the glucose tolerance of mice increased,and the blood lipid level decreased under the synergistic intervention of Momordica charantia herbal enzyme and metformin.Momordica charantia herbal enzyme has obvious auxiliary hypoglycemic effect,which lays a theoretical foundation for the development of hypoglycemic products.
