云南大百合与‘索蚌’百合体细胞融合培养初报 被引量:3

Preliminary Report on the Somatic Cell Fusion Culture of Cadiocrinum giganteum and Lilium oriental’Sorbonne’

摘要 本研究选用百合科百合属东方百合系‘索蚌’和百合科大百合属云南大百合的愈伤组织为材料,对其分离纯化的原生质体进行体细胞杂交及培养研究,拟克服远缘杂交障碍,为百合属间杂交和品种改良奠定基础。研究发现:比较3种不同浓度PEG及不同处理方法对原生质体融合的影响得出:改良PEG融合法最好,即A液(35%PEG),(27±3)℃,15 min,黑暗,融合率达30%。对融合后原生质体进行5种不同培养方法培养,结果表明:最适合融合原生质体培养方法为看护培养。在改良MS+Picloram 2 mg/L+6-BA 1.5 mg/L+NAA 0.1 mg/L+2,4-D 0.1 mg/L培养50 d后可获得再生细胞团,但未发育成愈伤组织。 In this research,Cadiocrinum giganteum and Lilium oriental’sorbonne’are used as material,somatic hybridization and culture of protoplasts isolated and purified from calli were studied in order to overcome the obstacles of distant hybridization and lay a foundation for intergeneric hybridization and variety improvement of lily.The results showed that:in experiment,compared with effects of three PEG concentration on protoplast fusion.The fusion condition was treated in dark conditions at(27±3)℃,for 15 minutes,35%PEG.The ratio of protoplast fusion is 30%.The results showed that the most suitable method for fusion protoplast culture was nursing culture.After improved MS+picloram 2 mg/L+6-BA 1.5 mg/L+NAA 0.1 mg/L+2,4-D 0.1 mg/L,50 days later regenerated cell clusters were obtained,but did not develop into callus.
