绿叶富贵竹组织培养再生体系的建立

Establishment of Tissue Culture Regeneration System of Dracaena sanderiana

摘要 为建立高效的绿叶富贵竹组织培养再生体系,以其茎段为外植体,针对消毒时间、增殖培养、生根培养、幼苗移栽等环节设置不同的实验处理开展研究。结果表明:绿叶富贵竹外植体最佳消毒处理方式为75%酒精预处理30 s,再经0.1%HgCl2消毒12 min,污染率为16.66%,芽萌生率为88.43%;适宜的基本培养基为MS,最佳的增殖培养基配方为MS+6-BA 3.5 mg/L+KT 1 mg/L+NAA 0.1 mg/L,增殖系数5.07;最佳的生根培养基配方为1/2MS+0.5 mg/L NAA,生根率达100%;幼苗移栽时,椰糠∶河沙∶珍珠岩按1∶1∶1的体积比为混合基质,移栽成活率达到96.67%,植株生长健壮。本研究成功建立了绿叶富贵竹组织培养再生体系,为其种苗快繁及遗传转化体系的建立奠定良好基础。 In order to establish an efficient tissue culture regeneration system of Dracaena sanderiana,the stems of D.sanderiana were used as explants in vitro,and different experimental treatments were set up for the suitable conditions of disinfection time,proliferation,rooting and transplanting were studied.The results showed that the best disinfection treatment was pretreatment was 75%alcohol for 30 s and 0.1%Hg Cl2for 12 min,with the lowest pollution rate of 16.66%and the germination rate was 88.43%.The selection of basic medium was MS.The best proliferation medium contained MS+6-BA 3.5 mg/L+KT 1 mg/L+NAA 0.1 mg/L,with the proliferation coefficient of5.07;The best rooting medium was consisted of 1/2MS+0.5 mg/L NAA,at a rooting rate could reach 100%;When seedlings were transplanted,the volume ratio of coconut coir∶fluvial sand∶pearlite was 1∶1∶1,the highest survival rate reached 96.67%,and the plant grew vigorously.This study successfully established a tissue culture regeneration system of D.sanderiana,which laid a foundation for the rapid propagation of seedlings and genetic transformation system in the future.
