摘要
目的观察藤龙补中汤对大肠癌荷瘤小鼠免疫功能的影响。方法将30只SPF级雄性BALB/c小鼠饲养1周后进行CT26细胞皮下注射,待肿瘤组织长至可测量范围时,按照随机数字表法分为对照组、复方斑蝥胶囊组和藤龙补中汤组,每组10只,分别予无菌水、复方斑蝥胶囊195 mg/kg和无菌水溶解藤龙补中汤提取物2.56 g/kg灌胃,每次0.2 mL,每日1次,共15 d。灌胃结束后取血、处死小鼠,称量脾、胸腺质量,计算脏器指数;流式细胞仪检测免疫细胞CD3+、CD4+、CD8+T淋巴细胞和自然杀伤(NK)细胞;酶标仪450 nm检测辅助型T淋巴细胞1(Th1)型细胞因子干扰素γ(IFN-γ)、白细胞介素12(IL-12)光密度,结果以对照组倍数表示。结果复方斑蝥胶囊组、藤龙补中汤组脾指数、胸腺指数均高于对照组(P<0.05);藤龙补中汤组胸腺指数高于复方斑蝥胶囊组(P<0.05);复方斑蝥胶囊组与藤龙补中汤组脾指数比较差异无统计学意义(P>0.05)。复方斑蝥胶囊组、藤龙补中汤组CD3+、CD4+、CD8+T淋巴细胞、NK细胞均高于对照组(P<0.05);藤龙补中汤组CD4+T淋巴细胞低于复方斑蝥胶囊组(P<0.05),藤龙补中汤组CD8+T淋巴细胞、NK细胞均高于复方斑蝥胶囊组(P<0.05);复方斑蝥胶囊组与藤龙补中汤组CD3+T淋巴细胞比较差异无统计学意义(P>0.05)。复方斑蝥胶囊组、藤龙补中汤组IFN-γ、IL-12均高于对照组(P<0.05);藤龙补中汤组IFN-γ、IL-12均高于复方斑蝥胶囊组(P<0.05)。结论藤龙补中汤可改善大肠癌荷瘤小鼠免疫功能,激发Th1型免疫反应。
Objective To observe the effects of Tenglong Buzhong Decoction(TLBZD)on immune function of tumor-bearing mice with colorectal cancer(CRC).Methods Totally 30 male BALB/C mice(SPF grade)fed 1 week were subcutaneously transplanted with CT26 cells.Until the tumor tissue grew to a measurable range,the mice were randomly assigned at 1∶1∶1 ratio to the control group(sterile water 0.2ml·d^(-1)),compound mylabris capsules group(CMC 195mg·kg^(-1)·d^(-1))and TLBZD group(sterile dissolved matter of TLBZD 2.56g·kg^(-1)·d^(-1)).After 15 days of treatment,blood drawn was taken,then the mice were sacrificed.The spleen and thymus were weighed,and the organ index was calculated.The immune cells(CD3^(+)、CD4^(+)、CD8^(+)T lymphocytes,natural killer[NK]cells)were detected using flow cytometry(FCM);the optical density(OD)of Th1 type cytokine(interferon-γ[IFN-γ],interleukin-12[IL-12])was measured with a microplate reader at 450 nm,and the results were expressed in multiples of the control group.Results The spleen index and thymus index were higher in CMC group and TLBZD group than in the control group(P<0.05);the thymus index was increased(P<0.05)in TLBZD group relevant to CMC group;none of the spleen index differed significantly between TLBZD group and CMC group(P>0.05).The immune cells in CMC group and TLBZD group were increased as compared with the control group(P<0.05);lower CD4^(+)T lymphocytes,and higher CD8^(+)T lymphocytes and NK cells were detected in TLBZD group than those in CMC group(all P<0.05);the difference was not statistically significant in the CD3^(+)T lymphocytes between TLBZD group and CMC group(P>0.05).IFN-γand IL-12 in the comparison of the three groups,TLBZD group was the highest,followed by CMC group and the control group(all P<0.05).Conclusion For tumor-bearing mice with CRC,TLBZD can improve immune function and stimulate Th1-type immune response.
