Effects of the activating transcription factor 2 on pancreatic cancer cells proliferation and apoptosis

目的观察激活转录因子2(ATF2)在胰腺癌细胞中的表达水平,探讨ATF2与胰腺癌细胞增殖和凋亡的关系。方法采用实时PCR和Western blotting检测14例胰腺癌和对应癌旁组织中ATF2 mRNA及蛋白的表达情况;构建ATF2 shRNA转染胰腺癌细胞株,检测转染后胰腺癌细胞的增殖及凋亡情况。结果在胰腺癌组织中ATF2 mRNA及蛋白表达水平高于癌旁组织(P<0.05),p-ATF2蛋白表达水平高于癌旁组织(P<0.01)。与对照组相比,ATF2 shRNA转染的胰腺癌细胞增殖被抑制,凋亡增加(P<0.01)。结论在胰腺癌细胞中ATF2表达水平增高,ATF2 shRNA转染胰腺癌细胞后,明显抑制细胞增殖,增加细胞凋亡。

Objective To evaluate the expression level of the activating transcription factor 2(ATF2)in pancreatic cancer(PC)cells,and to explore the relationship between ATF2 and PC cells proliferation and apoptosis.Methods Real-time PCR and Western blotting techniques were used to detect the expression of ATF2 messenger RNA(mRNA)and protein in 14 samples of PC tissues and adjacent normal tissues(ANT).ATF2 short hairpin RNA(shRNA)was constructed to transfect the PC cell lines,and cell proliferation and apoptosis were evaluated in the PC cells after the transfection.Results The expression levels of ATF2 mRNA and protein in the PC tissues were higher than in the ANT(P<0.05),and the expression level of phosphorylated ATF2(p-ATF2)protein was also higher in the PC tissues(P<0.01).Compared with the control group,the proliferation of PC cells transfected with ATF2 shRNA was inhibited,while apoptosis was increased(P<0.01).Conclusion The expression level of ATF2 is increased in PC cells.Transfection of PC cells with ATF2 shRNA can significantly inhibit cell proliferation and increase cell apoptosis.
