摘要
目的:从中医“同病异治”理论角度出发,探讨电针分别刺激足阳明胃经、督脉对脊髓损伤大鼠的损伤部位蛋白激酶A(PKA)的表达,进一步探讨电针刺激对脊髓损伤大鼠中枢神经轴突再生的机制。方法:将60只SPF级SD雌性大鼠随机分为对照组、督脉组与足阳明胃经组,每组大鼠分为1周、2周、3周、4周与5周共5个亚组,每个亚组4只大鼠。督脉组给予电针刺激阿是穴,足阳明胃经组给予电针刺激“足三里”“伏兔”穴,对照组仅损伤脊髓,不给予治疗。采用免疫组化、PCR与WB检测3组在各时间点阳性蛋白及蛋白激酶A的表达情况。结果:免疫组化结果显示电针刺激足阳明胃经、督脉对比对照组阳性蛋白表达明显增高,差异有统计学意义(P<0.05);电针刺激组的PKA的mRNA及蛋白表达较对照组显著升高(P<0.05),其中足阳明胃经组的阳性蛋白表达及PKA的mRNA及蛋白表达较督脉组高,在第3周时差异具有统计学意义,其他时间点差异无统计学意义。结论:电针刺激足阳明胃经、督脉穴可以促进脊髓损伤局部阳性蛋白及PKA的表达,符合中医“同病异治”理论,在促进轴突再生方面发挥重要作用。
Objective:To explore electro-needling Stomach Meridian and Du Meridian on the expression of PKA in the injured part of spinal cord injury(SCI)rats from the perspective of the TCM theory of‘same disease with different treatments’,and to further explore the mechanism of electro-acupuncture(EA)stimulation on central nerve axon regeneration in rats with SCI.Methods:60 SPF-level SD female rats were randomly divided into the control group,the Du Meridian group and the Stomach Meridian group.Each group was subdivided into subgroups of 1 w,2 w,3 w,4 w and 5 w,with 4 rats in each subgroup.The Du meridian group was treated with electro-needling Ashi points,the Stomach meridian group was treated with electro-needling Zusanli(ST36)and Futu(ST32),and no treatment intervention was given in the control group after SCI.Immunohistochemistry,PCR and WB were used to detect the expressions of positive protein and PKA in the three groups at each time point.Results:The results of immunohistochemistry showed that the positive protein expressions were significantly higher in the Stomach meridian group and the Du meridian group than those in the control group(P<0.05).The mRNA and protein expressions of PKA in the Du meridian group and the Stomach meridian were significantly higher than those of the control group(P<0.05);the positive protein expressions and the mRNA and protein expressions of PKA were higher in the Stomach meridian than those in the Du meridian group,which had significant significance at the 3rd week,and the difference was not significant at other time points.Conclusion:Electro-needling Stomach meridian and Du meridian can promote the expressions of local positive proteins and PKA in SCI,which conforms to the TCM theory of‘same disease with different treatments’,and plays an important role in promoting axon regeneration.
