摘要
目的:观察夹脊电针对兔退变腰椎间盘组织细胞凋亡的影响。方法:选取新西兰雄性大白兔20只,随机分为正常组、模型组、假模型组与电针组,每组5只。正常组给予正常喂养,不予任何处理;模型组在实验兔的L_(4)、L_(5)椎体间置入克氏针,连接加压装置,进行椎体间加压28 d,不做治疗;假模型组在实验兔的L_(4)、L_(5)椎体间置入克氏针,不进行椎体间加压,不做治疗;电针组在造模成功后,施以电针治疗28 d。各组行椎间盘MRI平扫,比较Pfrirmann椎间盘MRI分级,运用激光共聚焦方法观察Bax和Bcl-2蛋白表达,TUNEL染色法观察各组兔退变腰椎间盘组织细胞凋亡情况。结果:与正常组比较,假模型组椎间盘的MRI分级、Bax、Bcl-2表达及凋亡细胞阳性数的差异无统计学意义(P>0.05);模型组椎间盘的MRI分级较高,Bcl-2表达明显降低,Bax表达明显增加,凋亡细胞阳性数明显增多,差异有统计学意义(P<0.05)。与模型组比较,电针组椎间盘的MRI分级下降,Bax表达降低,Bcl-2表达增加,凋亡细胞阳性数减少,差异有统计学意义(P<0.05)。结论:电针夹脊穴能降低MRI分级指数,促进Bcl-2表达,抑制Bax表达,抑制椎间盘退变细胞凋亡。
Objective:To observe the effect of Jiaji electro-acupuncture(EA)on apoptosis of lumbar intervertebral disc tissues in rabbits.Methods:20 male New Zealand white rabbits were randomly divided into the normal group,the model group,the sham model group and the EA group,with 5 rabbits in each group.The normal group was given normal feeding without any other intervention.In the model group,Kirschner wire was inserted between L4 and L5 vertebrae,and a compression device was connected to perform intervertebral compression for 28 days without treatment.In the sham model group,Kirschner wire was inserted between L4 and L5 vertebrae without intervertebral compression or treatment.The EA group was treated with EA for 28 days after successful modeling.The MRI of intervertebral disc was performed in each group,and the MRI grading of Pfrirmann intervertebral disc was compared.The protein expressions of Bax and Bcl-2 were observed by laser confocal method,and the apoptosis of lumbar intervertebral disc tissues was observed by TUNEL staining.Results:There were no statistical differences in MRI grading,expressions of Bax and Bcl-2 and positive number of apoptotic cells between the normal group and the sham model group(P>0.05).Compared with those in the normal group,the MRI grading of intervertebral disc was higher,the expression of Bcl-2 was significantly decreased,the expression of Bax was significantly increased,and the positive number of apoptotic cells was significantly increased in the model group(P<0.05).Compared with those in the model group,the MRI grading of the intervertebral disc was decreased,the expression of Bax was decreased,the expression of Bcl-2 was increased,and the positive number of apoptotic cells was decreased in the EA group,and the differences were statistically significant(P<0.05).Conclusion:Electro-needling Jiaji points can reduce MRI grading indexes,promote the expression of Bcl-2,inhibit the expression of Bax and inhibit the apoptosis of intervertebral disc degeneration cells.
