摘要
目的:研究穴位电刺激对卒中模型大鼠认知功能的影响及可能作用机制。方法:将36只SPF级雄性SD大鼠随机分为假手术组10只与手术组26只。手术组采用改良Longa线栓法建立大脑中动脉缺血(MCAO)模型;假手术组仅作颈动脉分离,不进行造模。大鼠完全清醒后,按照Zea Longa评分法将手术组1~3分大鼠随机纳入模型组与治疗组。造模后1 d对治疗组大鼠神庭穴、百会穴施以连续2周的电针治疗。模型组与假手术组大鼠只固定时间抓取,不给予任何干预措施。治疗结束后,采用Morris水迷宫实验检测各组大鼠认知功能,取大鼠脑组织检测脑组织含水量,酶联免疫吸附测定(ELISA)法检测大鼠脑组织超氧化物歧化酶(SOD)及丙二醛(MDA)含量,qPCR法检测大鼠脑组织PI3k和Akt mRNA表达水平。结果:模型组、治疗组与假手术组逃避潜伏期依次缩短,目标象限停留时间依次延长,穿越原平台次数依次增加,脑组织含水量依次减少,脑组织SOD含量依次增高,MDA含量依次降低,差异具有统计学意义(P<0.05);治疗组PI3k、Akt mRNA表达水平明显高于假手术组及模型组。结论:电针干预百会穴、神庭穴可改善脑卒中大鼠认知功能,缓解脑水肿,其作用机制可能与改善脑组织SOD活性、减轻脑组织MDA蓄积和上调PI3k/Akt通路表达有关。
Objective:To investigate the effect and possible mechanism of electrical stimulation to acupoints on cognitive function in stroke model rats.Methods:36 male SD rats of SPF grade were randomly divided into the sham operation group(n=10)and the operation group(n=26).The operation group was established the model of middle cerebral artery ischemia(MCAO)with modified Longa line suppository method,whereas the sham operation group was only performed carotid artery separation.After the rats were fully awake,rats with 1~3 points in the operation group were randomly assigned into the model group and the treatment group according to the Zea Longa scoring method.1 day after modeling,the treatment group was treated with electro-needling Shenting(DU24)and Baihui(DU20)for 2 consecutive weeks.Rats in the model group and the sham operation group were only held at a fixed time and no intervention was given.After the treatment,the Morris water maze experiment was used to detect the cognitive function of the rats in each group,the brain tissue of the rats was taken to detect the water content of brain tissue,the contents of SOD and MDA of rat brain tissue were detected by ELISA,and the mRNA expressions of PI3k and Akt of rat brain tissue were detected by qPCR.Results:The latency period of evasion was shortened sequentially(P<0.05),the dwell time of the target quadrant was extended sequentially(P<0.05),the number of crossings through the original plateau increased sequentially(P<0.05),the water content of brain tissue decreased sequentially(P<0.05),the SOD content of brain tissue increased sequentially(P<0.05),and the MDA content of brain tissue decreased sequentially in the model group,the treatment group and the sham operation group(P<0.05).The mRNA expressions of PI3k and Akt were significantly higher in the treatment group than those in the sham operation group and the model group(P<0.05).Conclusion:Electro-needling DU20 and DU24 can improve the cognitive function and alleviate cerebral edema in stroke rats,and its action mechanism may be related to improving SOD activity of brain tissue,alleviating the accumulation of MDA in brain tissue,and up-regulating the pathway expression of PI3k/Akt.
