秦芋32号马铃薯花药培养技术研究

摘要 研究使用陕西省安康市主载马铃薯品种“秦芋32号”为材料,研究外植体消毒方式、培养基附加物、培养方式等对花药膨大以及褐化的影响。结果表明,选择花药瓣药比0.75~1.0,花药黄绿色,此时处于单核靠边期的花粉概率较高,适合用作花药组织培养,酒精+升汞消毒能有效的减轻秦芋32号花药的褐化,培养基中加入适量肌醇,有助于提高秦芋32号花药的膨大率,并降低花药褐化率,培养基中添加适量甘氨酸,对促进花药膨大有显著作用,但也加剧了花药的褐化现象,可结合酒精+升汞消毒降低褐化影响,同时,光照培养能有效促进秦芋32号花药的膨大。综合以上研究:选择瓣药比0.75~1.0的黄绿色花药,外植体采用酒精+升汞消毒,培养基添加5~10 mg/L的肌醇,50mg/L的甘氨酸,愈伤组织选择光照培养为秦芋32号花药培养的适宜条件。 The main potato variety‘Qinyu 32’in Ankang was used as experimental material to study the effects of explant disinfection,medium appendages and culture methods on anther expansion and browning.The results showed that when the ratio of anther petal to anther was 0.75~1.0,the anther was yellowish green,and the pollen at the uninucleate stage had a higher probability,which was suitable for anther tissue culture.Alcohol and mercuric disinfection could effectively reduce the browning of‘Qinyu 32’anther.Addition of an appropriate amount of inositol to the culture medium could help improve the swelling rate of‘Qinyu 32’anther and reduce the browning rate of anther.Addition of an appropriate amount of glycine to the culture medium could significantly promote anther swelling,but also aggravate the browning phenomenon of anther,which could be combined with alcohol and mercuric disinfection to reduce the browning effect.At the same time,Light culture could effectively promote the expansion of anther‘Qinyu 32’.Based on the above studies:the yellow green anther with the valve drug ratio of 0.75-1.0 was selected,the explants were disinfected with alcohol and mercuric chloride,the medium was added with 5mg/L-10mg/L inositol and 50mg/L glycine,and the light culture of callus was selected to be the suitable condition for anther culture of‘Qinyu 32’.
