摘要 基因调节受损是许多疾病的核心,包括发育系统疾病和癌症。N6甲基腺苷的可调节的甲基化是哺乳动物RNA修饰的重要组成部分,已被越来越多的研究证明其在各种生物学过程及癌症发病机制中发挥重要作用。这种RNA化学标记可由“writers”蛋白(甲基转移酶)产生,并可由“ereasers”蛋白(去甲基酶)逆转,这些化学修饰可以由“readers”蛋白(甲基化阅读蛋白)识别,并依此来调节下游分子机制。RNA m^(6)A修饰在重要的生物学过程中起着重要作用,有研究表明,m^(6)A在NSCLC的发生、转移及侵袭方面至关重要,m^(6)A的研究也为NSCLC治疗提供了新的思路,本文就m^(6)A在NSCLC中的病因、发展、耐药等方面进行一一论述,目的是从表观遗传学方面进一步挖掘肺癌发生发展的生物学机制,为NSCLC的靶向治疗提供研究思路。 Impaired gene regulation lies at the core of many diseases,including developmental system diseases and cancer.The adjustable N-6 methyladenosine(m^(6)A)is an important part of mammalian RNA modification,and more and more studies have proved that it plays an important role in various biological processes and cancer pathogenesis.This RNA chemical marker can be made of the"writers"protein(methyltransferases)and can be reversed by the"ereasers"protein(demethylases).These chemical modifications can be recognized by the"readers"protein(m^(6)A binding proteins).And adjust the downstream molecular mechanism.RNA m^(6)A modification plays an important role in important biological processes.Some studies have shown that m^(6)A plays an important role in the occurrence,metastasis and invasion of NSCLC.The research of m^(6)A also provides new ideas for the treatment of NSCLC.The causes,development,drug resistance and treatment of lung cancer are discussed one by one.The purpose is to further explore the pathogenesis of lung cancer from the perspective of epigenetics and provide research direction for targeting therapy of NSCLC.
