摘要
本研究以玫瑰茄为材料,采用乙醇-(NH)SO双水相系统提取玫瑰茄活性成分花色苷。根据单因子测试结果,进行响应面分析,优化花色苷的提取。通过体外抗氧化活性实验,研究了最佳提取条件下玫瑰茄提取物的羟基自由基清除率。结果显示,乙醇体积分数为40%,(NH)SO质量浓度为0.24g/mL,液料比为40∶1(mL/g),40.5℃,24.5min条件下,花色苷提取量最高为4.121mg/g,与预测值接近;玫瑰茄花色苷提取物清除羟自由基的IC值为169.1μg/mL,维生素C的IC_(50)值为448.5μg/mL,花色苷提取物总抗氧化能力优于维生素C。本研究结果有助于实现玫瑰茄资源的高效利用。
In this study,the anthocyanins of roselle were extracted by ethanol-(NH)SOaqueous two-phase system.According to the results of single factor test,Response surface analysis was formulated and the extraction process of anthocyanins was optimized.The antioxidant activity experiment in vitro was carried out to study the hydroxyl radical scavenging rate of roselle extract under the optimized extraction process.The results showed that when the volume fraction of ethanol was 40%,the mass concentration of(NH)SOwas 0.24g/mL,when the liquid-solid ratio was 40∶1(mL/g),at 40.5℃,for 24.5min,the extraction amount of anthocyanin was 4.121mg/g,which was close to the predicted value;The ICvalue of roselle anthocyanin extract for scavenging hydroxyl radical was 169.1μg/mL,the IC_(50)value of vitamin C was 448.5μg/mL,the total antioxidant capacity of anthocyanin extract was better than that of vitamin C.This study is helpful to realize the efficient utilization of roselle resources.
