摘要
阿尔茨海默病(Alzheimer′s disease,AD)是一种中枢神经退行性疾病,多发于老年人.细胞内钙离子(Ca^(2+))是第二信使分子,其参与了β-淀粉样蛋白产生、Tau蛋白过度磷酸化、早老素突变、突触功能障碍和线粒体功能障碍等AD发病的全过程.本文分别从质膜和细胞器钙离子信号通路方面对AD患者脑内钙离子调节异常的分子机制以及相关代表性的药物进行了综述,以期更好地对AD进行防治.
Alzheimer′s disease(AD)is a degenerative disease of the central nervous system,which mainly affects the elderly.Intracellular calcium(Ca^(2+))is a second messenger molecule,which is involved in the whole process of AD,such as amyloid-βproduction,Tau hyperphosphorylation,presenilin mutation,synaptic dysfunctionand mitochondrial dysfunction.This article summarizes the molecular mechanism of abnormal calcium regulation in the brain of patients with AD from the aspects of plasma membrane and organelle calcium signaling pathway,as well as the representative drugs,in order to better prevent and treat AD.
