金枝黄花柳回交子代枝条颜色与转录组研究 被引量:1

摘要 为了探究枝条颜色变异的原因,以金枝黄花柳回交子代及其亲本为材料,分析子代枝条性状遗传变异规律及性状与基因表达的关系。结果显示,金枝型回交子代枝条表现为黄色到橙色,普通型回交子代枝条表现为墨绿色至棕色,不同单株个体之间存在颜色差异。枝条转录组共检测到差异表达基因2499个,其中上调基因984个,下调基因1515个。功能注释和富集分析发现,卟啉与叶绿素代谢通路(KEGG&ko00860)中与色素合成有关的基因表达量存在差异,其中叶绿素/细菌叶绿素a合成酶(2.5.1.133;2.5.1.62)与叶绿素/细菌叶绿素a还原酶(1.3.1.111)基因表达上调,叶绿素酶(3.1.1.14)基因表达下调。基因差异表达会影响代谢通路,进而影响枝条中叶绿素的含量,导致枝条颜色存在差异。 In order to explore the cause of variation in branch color,this study analyzed the genetic variation of branch traits and the relationship between traits and gene expression in the backcross progenies of Salix caprea var.aurea and their parents.The results showed that the branches of the golden type backcross progeny were yellow to orange,and the branches of the common type backcross progeny were dark green to brown,and there were color differences among individual plants.A total of 2499 differentially expressed genes were detected,including 984 up-regulated genes and 1515 down-regulated genes.Functional annotation and enrichment analysis showed that the expression levels of genes related to pigment synthesis were different between porphyrin and chlorophyll metabolic pathway(KEGG&ko00860).Chlorophyll/bacterial chlorophyll A synthase(2.5.1.133;2.5.1.62)and chlorophyll/bacterial chlorophyll a reductase(1.3.1.111)gene expression was up-regulated,and chlorophyllase(3.1.1.14)gene expression was down-regulated.Differential gene expression could affect metabolic pathways,and then affect chlorophyll content in branches,resulting in differences in branches color.
