多囊卵巢综合征痰湿证患者血清差异蛋白质组学研究

Study on serum differential proteomics in patients with polycystic ovary syndrome with phlegm dampness syndrome

目的应用蛋白质组学探讨多囊卵巢综合征(PCOS)的潜在血清特异生物标志物。方法分别采集2019年4月—2020年10月于山西省中西医结合医院和黑龙江中医药大学附属第一医院的12例PCOS痰湿证患者和同期健康对照者血清,采用相对和绝对定量同位素标记技术(iTRAQ)联合超高液相色谱串联质谱(LC-MS/MS)进行蛋白组学分析,寻找差异表达的蛋白质。结果经质谱分析共得到有意义差异蛋白646个,筛选出87种显著性差异蛋白(P<0.05),其中胰岛素样生长因子Ⅱ、胱抑素-C等10种蛋白表达上调,补体C3、载脂蛋白B、维生素D结合蛋白等77种蛋白表达下调。这些差异蛋白共涉及8条KEGG信号/代谢通路。结论应用iTRAQ联合LC-MS/MS筛选出了PCOS痰湿证的血清差异表达蛋白,为寻找PCOS痰湿证特异性标志物提供了生物学基础。

Objective It is to explore the potential serum specific biomarkers of polycystic ovary syndrome(PCOS)with proteomics.Methods The serum of 12 PCOS patients with phlegm dampness syndrome and healthy controls treated in Shanxi Provincial Hospital of Integrated Traditional Chinese and Western Medicine and the First Affiliated Hospital of Heilongjiang University of Traditional Chinese Medicine from April 2019 to October 2020 were collected for proteomic analysis by isobaric tag for relative absolute quantitation(iTRAQ)combined with liquid chromatography tandem mass spectrometry(LC-MS/MS)to find differentially expressed proteins.Results A total of 646 significant differential proteins were obtained by mass spectrometry analysis,and 87 significant differential proteins were screened(P<0.05).Among them,10 proteins such as insulin-like growth factor II and cystatin C were up-regulated,while 77 proteins such as complement C3,apolipoprotein B and vitamin D binding protein were down-regulated.These differential proteins totally involved 8 KEGG signaling/metabolic pathways.Conclusion Using iTRAQ combined with LC-MS/MS,we screened the serum differential expression proteins of PCOS with phlegm dampness syndrome,which provides a biological basis for searching for specific markers of PCOS with phlegm dampness syndrome.
