基于微卫星复合扩增的马来穿山甲个体识别研究

摘要 利用12个微卫星位点复合扩增构建马来穿山甲(Manis javanica)个体识别体系,评估其种群遗传多样性和遗传结构。结果显示:重构的个体识别体系累积无关个体基因型匹配概率(P_((ID)))和同胞个体基因型匹配概率(P_((ID)sibs))分别为6.48×10^(-16)和9.65×10^(-6),具有较高的鉴别力和灵敏度。在135只马来穿山甲个体上共检测出155个等位基因,多态信息含量(PIC)为0.733,平均等位基因数(N_(a))为12.91,平均观测杂合度(H_(o))为0.631,平均期望杂合度(H_(e))为0.756,在鲮鲤科(Manidae)内处于高水平。聚类分析显示样本集分为3个种群,具有显著的遗传结构。重构的马来穿山甲个体识别体系具有可靠性强、成本低、物种特异性,以及准确率和灵敏度高等优点,适用于多种类型检材。同时,该体系可用于评估马来穿山甲种群遗传多样性水平和遗传结构。 In this study,an individual identification system of Malayan pangolin(Manis javanica)was constructed by compound amplification of 12 microsatellite locus,and the genetic diversity and structure of involved population were evaluated.The results showed that the cumulative unrelated individual genotype matching probability(P_((ID)))and sibling genotype matching probability(P_((ID)sibs))of the reconstructed individual identification system were 6.48×10^(-16) and 9.65×10^(-6) respectively,indicating high discriminative power and sensitivity.A total of 155 alleles were detected in 135 Malayan pangolin individuals,the polymorphic information content(PIC)was 0.733,the mean number of alleles(N_(a))was 12.91,the mean observed heterozygosity(H_(o))and mean expected heterozygosity(H_(e))were 0.631 and 0.756 respectively,which were at higher levels within the Manidae.Cluster analysis showed that these samples were divided into three populations with significant genetic structure.The reconstructed Malayan pangolin identification system has strong reliability,species specificity,high accuracy and sensitivity,low cost,and can be adapted to various types of samples.Meantime,this system can be used to assess the genetic diversity and structure for Malayan pangolin populations.
