摘要
目的:探讨结直肠癌肝转移瘤中肝脏远癌组织的亚细胞结构与表达谱改变情况。方法:电镜观察人体肝脏远癌组织亚细胞结构变化,通过建立动物结直肠癌肝转移模型观察小鼠远癌肝组织亚细胞结构变化;利用转录组测序探究动物模型中肝脏远癌组织的表达谱改变。结果:人与小鼠肝转移组织中的亚细胞结构均发生异于正常组织的改变。转录组分析结果显示在远癌组织中上调基因的表达和富集通路均与肿瘤发生发展相关。结论:远癌组织的形态结构与基因表达发生类似于肿瘤组织的改变。
Objective:To investigate the changes of subcellular structure and expression profile of distal tissue in colorectal cancer liver metastasis.Methods:The structural changes of human liver were observed by electron microscope,and confirmed in a colorectal cancer liver metastasis mice model.Transcriptome profiling was used to explore the differential expressed genes in distal tissues in the mice model.Results:The subcellular structure of liver cells in distant tissue in both human and mouse liver metastases were obviously different from that of healthy liver cells.Transcriptome analysis showed that differentiated genes in distal tissues were enriched to pathways related to tumor.Conclusion:The morphological structure and gene expression of distal tissues were also affected by the liver metastasis.
