摘要 目的甘油三酯(TG)作为主要的储备能源,是一种高度压缩的能源,它储存的能量相当于同重量水化糖原的6倍之多。而它也有一个缺点:动用时不像糖那么方便。甘油三脂的合成及分解途径分为两种:人体内途径及体外生物工程途径。方法主要对比32例不同人血浆中血脂四项[TG、总胆固醇(TC)、高密度脂蛋白(HDL)及低密度脂蛋白(LDL)]的指标差异、5种微生物脂肪酶效价及分解甘油三酯的效果,并综合相关文献报道,比较微生物脂肪酶的优劣。结果32例样本中筛选出23例高甘油三脂样本,但是这些样本中TC、HDL及LDL均在正常值范围,且高甘油三脂样本中的TC、HDL及LDL含量并不高。TG与TC、HDL及LDL无显著相关性。不同脂肪酶效价差别很大,比较5种脂肪酶效价(使用活性)。脂肪酶1效果最好。脂肪酶1处理高血脂样本50 u/mL与100 u/mL无显著差异,选择50 u/mL比较合适。结论本文通过高血脂样本的筛选,5种不同脂肪酶的酶活测定及脂解高血脂样本的效果,评价筛选出1种脂肪酶可用于高血脂样本的体外酶解,改善体外诊断试剂中高血脂样本的干扰,提高检测准确性。 【Objective】Triglyceride(TG),as the main reserve energy,is a highly compressed energy,which stores up to 6 times as much energy as hydrated glycogen by weight.And it has one drawback:it's not as easy to handle as sugar.The synthesis and decomposition of triglycerides can be divided into two pathways:in vivo and in vitro bioengineering.【Methods】In this study,the differences of four blood lipids[TG(triglyceride),TC(total cholesterol),HDL(high density lipoprotein)and LDL(low density lipoprotein)],the titer of five microbial lipases and the effect of triglyceride decomposition were compared in 32 cases of different human plasma,and the advantages and disadvantages of microbial lipases were compared based on relevant literature reports.【Results】Twenty-three cases of high triglyceride samples were screened out from 32 samples,but the TC,HDL and LDL in these samples were within the normal range,and the contents of TC,HDL and LDL in these samples were not high.There was no significant correlation between TG and TC,HDL and LDL(P<0.05).Different lipase titers were very different.Five lipase titers(using activity)were compared.Lipase 1 worked best.Lipase 1 treatment of hyperlipidemia samples 50 u/mL and 100 u/mL showed no significant difference,50 u/mL is more appropriate.【Conclusion】In this paper,through the screening of hyperlipidemia samples,the enzyme activity determination of 5 different lipases and the effect of lipolysis of hyperlipidemia samples,one lipase can be used for in vitro enzymatic hydrolysis of hyperlipidemia samples to improve the interference of hyperlipidemia samples in vitro diagnostic reagents and improve the accuracy of detection.
