摘要
糖尿病是一种以血糖持续升高为特征的常见代谢性疾病,其发病的关键原因之一是胰岛β细胞功能衰竭。硫氧还蛋白相互作用蛋白(TXNIP)可以结合并抑制葡萄糖摄入后胰岛产生的硫氧还蛋白,导致胰岛β细胞抗氧化能力降低。大量研究表明,TXNIP水平升高可使胰岛β细胞凋亡,反之,TXNIP水平下降则可显著促进胰岛β细胞存活。靶向TXNIP不仅可以改善胰岛β细胞胰岛素的分泌和敏感性,还能降低长期罹患糖尿病而导致的多种并发症。因此,调节TXNIP水平可作为降低糖尿病及其并发症发生的新型治疗策略。
Diabetes is a common metabolic disease characterized by continuous elevation of blood glucose.One of the key causes of diabetes is islet β cell failure.Thioredoxin Interacting Protein(TXNIP) can bind and inhibit the thioredoxin produced by islets after glucose intake,resulting in a decreased antioxidant capacity of β cells.Many studies show that the increase of TXNIP level can make pancreatic islets β cell apoptosis.On the contrary,the decrease of TXNIP level can significantly promote islet β cell survival.Targeting TXNIP can not only improve the insulin secretion and sensitivity of islets β cells,but also can reduce many complications caused by chronic diabetes.Therefore,regulating the level of TXNIP can be used as a new therapeutic strategy to reduce the incidence of diabetes and its complications.
