黄芩汤对人肝癌细胞PTEN、HIF-1α、VEGF表达的影响及机制研究 被引量:2

摘要 目的探讨黄芩汤对人肝癌HepG2细胞PTEN、HIF-1α、VEGF表达的影响及潜在机制。方法将人肝癌HepG2细胞分为空白组、对照组、黄芩汤低剂量组、黄芩汤中剂量组、黄芩汤高剂量组,分别给予培养液、2μmol/L浓度的5-氟尿嘧啶、0.5 mg/mL浓度黄芩汤、2.5 mg/mL浓度黄芩汤、5 mg/mL浓度黄芩汤。应用Western blot法、免疫细胞化学法检测PTEN、HIF-1α、VEGF蛋白表达。结果Western blot显示,5-氟尿嘧啶、黄芩汤作用24 h后,各干预组肝癌HepG2细胞的PTEN升高,HIF-1α、VEGF蛋白表达下降,与空白组比较差异有统计学意义(P<0.05)。干预后,黄芩汤高剂量组肝癌HepG2细胞PTEN表达量最高,HIF-1α、VEGF水平最低,与其他各组比较,差异有统计学意义(P<0.05)。结论黄芩汤能够提高人肝癌HepG2细胞的PTEN表达,抑制HIF-1α、VEGF蛋白表达,且呈剂量依赖性表达,这为临床治疗肝癌提供了新的思路。 Objective To investigate the effect of Huangqin decoction on expression of PTEN,HIF-1αand VEGF in HepG2 cells and its potential mechanism.Methods HepG2 cells were divided into blank group,control group,low-dose group,medium-dose group and high-dose group.The cells were given culture medium,2μmol/L 5-fluorouracil,0.5 mg/mL,2.5 mg/mL and 5 mg/mL of Huangqin decoction decoction,respectively.The expression of PTEN,HIF-1αand VEGF were detected by Western blot and immunocytochemistry.Results After treatment,western blot showed that with 5-fluorouracil and Huangqin decoction for 24h,PTEN of hepatocellular carcinoma HepG2 cells in each intervention group increased,HIF-1αand VEGF protein expression decreased,compared with the blank group difference statistically significant(P<0.05).After intervention,the expression level of PTEN in HepG2 cells was the highest and the level of HIF-1αand VEGF was the lowest in Huangqin decoction high-dose group,compared with other groups difference statistically significant(P<0.05).Conclusion Huangqin decoction can increase the expression of PTEN and inhibit the expression of HIF-1αand VEGF in dose-dependent manner in HepG2 cells.This provides new idea for clinical treatment of liver cancer.
