霍山铁皮石斛多糖脂质体的制备及质量评价

Preparation and Quality Evaluation of Polysaccharide Liposome of Dendrobii Officinalis Huoshanense

摘要 目的探讨霍山铁皮石斛多糖脂质体的制备工艺,并对其质量进行评价。方法采用薄膜分散超声法制备霍山铁皮石斛多糖脂质体,并以包封率为评价指标,采用单因素试验及正交试验筛选出霍山铁皮石斛多糖脂质体的最优制备工艺,并同时测定脂质体的粒径及电位。结果霍山铁皮石斛多糖脂质体的最佳制备工艺为大豆卵磷脂与药物比为20∶1,大豆卵磷脂与胆固醇比为10∶1,氯仿与甲醇比为1∶1,超声时间10 min,包封率为54.62%,平均粒径为183.5 nm,电位为-33.6 mV,体外12 h累积释药率40%。结论制备的霍山铁皮石斛多糖脂质体具有较高的包封率及较明显的缓释效应,粒径和形态较均匀,重复性好。 Objective To explore the preparation process of polysaccharide liposomes of Dendrobii Officinalis Huoshanense,and evaluate its quality.Methods The thin film dispersion-ultrasonic method was used to prepare polysaccharide liposome of Dendrobii Officinalis Huoshanense,and the encapsulation rate was taken as an evaluation index.Single factor test and orthogonal experiment were used to screen out the optimal preparation process of polysaccharide liposome of Dendrobii Officinalis Huoshanense,and the particle size and potentials of the liposomes were measured simultaneously.Results The optimal preparation process was as follows:Soybean Lecithin-drug ratio of 20∶1,chloroform to methanol ratio was 1∶1,ultrasonic time of 10 min,the encapsulation rate was 54.62%,average particle size of 183.5 nm and potentials of-33.6 mV.The cumulative drug release rate in vitro for 12 h was 40%.Conclusion The prepared polysaccharide liposomes of Dendrobii Officinalis Huoshanense has a high encapsulation rate and obvious slow release effect.The particle size and morphology of liposomes were uniform and reproducibility.
