子痫前期miR-30c-5p和Notch1的表达及相关性 被引量:1

摘要 目的探讨miR-30c-5p和Notch1在子痫前期(PE)患者胎盘中的表达及其相关性。方法选取中国人民解放军联勤保障部队第九六〇医院产科2021年1月至12月行剖宫产分娩的产妇60例,正常妊娠组20例,中度PE组20例,重度PE组20例。采用荧光定量PCR和免疫组织化学分别检测胎盘组织中miR-30c-5p和Notch1的表达水平;采用SPSS 25.0统计学软件分析数据;采用Pearson相关性分析检测miR-30c-5p与Notch1的相关性;采用ROC曲线分析miR-30c-5p与Notch1对疾病诊断的特异性。结果与正常妊娠组比较,中度PE组和重度PE组miR-30c-5p表达水平均显著升高(P<0.05),重度PE组miR-30c-5p表达水平较中度PE组有升高趋势。与正常妊娠组比较,中度PE组和重度PE组胎盘组织中Notch1表达水平均显著降低(P<0.05);且PE越重,胎盘组织中Notch1表达水平越低。miR-30c-5p与Notch1的表达呈明显负相关(r=-0.484,P<0.001)。miR-30c-5p诊断PE的AUC为0.625,诊断性能较好,但差异无统计学意义;Notch1诊断PE的AUC为0.914,诊断效能为优,其表达值对PE的诊断有意义。结论胎盘滋养细胞中miR-30c-5p负向调控靶基因Notch1,从而抑制滋养细胞的增殖、浸润,导致胎盘灌注不足,从而参与并加重PE的发生。 Objective To investigate expression and relevance of miR-30c-5p and Notch1 in placenta of patients with preeclampsia(PE).Methods A total of 60 delivery by cesarean section patients treated in the Department of Obstetrics and Gynecology of The 960th Hospital of Joint Logistics Support Force of PLA from January to Decenmber 2021 were selected and divided into the normal pregnancy group,the moderate PE group and the severe PE group,with 20 cases in each group.The quantitative real-time PCR(qPCR)and immunohistochemistry were used to detect the expression level of miR-30c-5p and Notch1 in the issue of placenta.SPSS 25.0 statistical software was used to analyze the data.The Pearson correlation analysis was used to detect the relevance between miR-30c-5p and Notch1.The receiver operating characteristic(ROC)curve was used to analyze the specificity for disease diagnosis of miR-30c-5p and Notch1.Results Compared with the normal pregnancy group,the expression level of miR-30c-5p in the moderate group and the severe PE group were significantly increased(P<0.05),while compared with the moderate PE group,the expression level of miR-30c-5p in the severe PE group increased.Compared with the normal pregnancy group,the expression level of Notch1 in the moderate PE group and the severe PE group were significantly decreased(P<0.05).After treatment,the clinical efficacy of the control group was significantly better than the study group,while there was a significant difference in clinical indexes after surgery between the two groups(P<0.05),while the severer the PE,the lower the expression level of Notch1 in placena.The expression of miR-30c-5p and Notch1 has a significant negative correlation(r=-0.484,P<0.001).The AUC of PE diagnosed by miR-30c-5p was 0.625,which had a good diagnostic performance but without significance.The AUC of PE diagnosed by Notch1 was 0.914,which had an excellent diagnostic performance,while the expression level of Notch1 was significant for diagnosis of PE.Conclusion miR-30c-5p has a negative regulation to target gene Notch1 in Placental trophoblastic cells,which can inhibit the proliferation and infiltration of trophoblastic cells,make the hypoperfusion of placenta and participate in and aggravate the degree of PE.
