慢性阻塞性肺疾病患者外周血单个核细胞中S100A10表达及意义

摘要 目的探讨S100A10在慢性阻塞性肺疾病(COPD)患者外周血单个核细胞(PBMC)中的表达变化及意义。方法选择2020年8月至2021年6月新乡医学院附属人民医院收治的42例COPD患者(COPD组)和同期体检健康的31例志愿者(健康对照组)为研究对象,比较2组受试者第1秒用力呼气容积(FEV_(1))占预计值的百分比(FEV_(1)%pred)和FEV_(1)占用力肺活量(FVC)的百分比(FEV_(1)/FVC);应用实时荧光定量聚合酶链反应法检测2组受试者PBMC中S100A10 mRNA相对表达量,Western blot法检测2组受试者PBMC中S100A10蛋白相对表达量,流式细胞术检测2组受试者PBMC各细胞亚群中S100A10蛋白相对表达量,分析COPD患者PBMC中S100A10 mRNA表达水平与FEV_(1)%pred的相关性。结果COPD组患者的FEV_(1)%pred、FEV_(1)/FVC显著低于健康对照组(P<0.05)。COPD组患者PBMC中S100A10 mRNA和蛋白相对表达量显著低于健康对照组(P<0.05)。COPD组与健康对照组受试者CD4^(+)T淋巴细胞、CD8^(+)T淋巴细胞、CD19+B淋巴细胞中S100A10蛋白相对表达量比较差异无统计学意义(P>0.05);COPD组患者的CD14^(+)单核细胞中S100A10蛋白相对表达量表达显著低于健康对照组(P<0.05)。COPD组患者PBMC中S100A10 mRNA表达水平与FEV_(1)%pred呈负相关(r=-0.610,P<0.05)。结论COPD患者PBMC中S100A10表达水平显著降低,其表达水平与FEV_(1)%pred呈负相关,S100A10可能在COPD发生发展中发挥重要调节作用。 Objective To investigate the change of expression of S100A10 in peripheral blood mononuclear cell(PBMC)of patients with chronic obstructive pulmonary disease(COPD)and its significance.Methods A total of 42 patients with COPD(COPD group)and 31 healthy volunteers(healthy control group)admitted to the Affiliated Renmin Hospital of Xinxiang Medical University from August 2020 to June 2021 were selected as the study objects.The percentage of forced expiratory volume in the first second(FEV_(1)%pred)and the percentage of FEV_(1) to forced vital capacity(FEV_(1)/FVC)of the subjects in the two groups were compared;the relative expression of S100A10 mRNA in PBMC of the subjects in the two groups were detected by real time fluorescent quantitative polymerase chain reaction,the relative expression of S100A10 protein in PBMC of the subjects in the two groups were detected by Western blot,and the relative expression of S100A10 protein in each cell subpopulation of PBMC of the subjects in the two groups was detected by flow cytometry.The correlation between the expression level of S100A10 mRNA in PBMC and FEV_(1)%pred of COPD patients was analyzed.Results FEV_(1)%pred and FEV_(1)/FVC of patients in the COPD group were significantly lower than those in the healthy control group(P<0.05).The relative expressions of S100A10 mRNA and protein in PBMC of patients in the COPD group was significantly lower than those in the healthy control group(P<0.05).There was on significant difference in the relative expression of S100A10 protein in CD4^(+)T lymphocytes,CD8^(+)T lymphocytes and CD19+B lymphocytes of the subjects between the COPD group and healthy control group(P>0.05);the relative expression of S100A10 protein in CD14^(+)monocytes of patients in the COPD group was significantly lower than that in the healthy control group(P<0.05).The expression level of S100A10 mRNA in PBMC was negatively correlated with FEV_(1)%pred of COPD patients(r=-0.610,P<0.05).Conclusion The expression level of S100A10 in PBMC of COPD patients was significantly decreased,and its expression level was negatively correlated with FEV_(1)%pred;S100A10 may play an important regulatory role in the development of COPD。
