摘要
三叶草黄脉病毒(Clover yellow vein virus,ClYVV)是马铃薯Y病毒属的重要成员,侵染多种豆科作物并造成严重危害。为明确从江苏省盐城市采集的蚕豆样品中检测出的ClYVV全基因组序列以及基因组结构特征,通过分段扩增的策略对ClYVV全基因组序列进行克隆,序列测定后拼接获得江苏蚕豆三叶草黄脉病毒(ClYVV-JS)全基因组序列。基因组序列分析结果显示该分离物基因组序列全长9585 bp,编码11个蛋白质。NCBI序列比对结果表明该分离物与已报道的日本ClYVV No.30分离物(AB011819)核苷酸序列同源性最高,达96.37%,其中编码的氨基酸序列同源性高达99.06%,而与中国合肥ClYVV分离物(KU922565)核苷酸序列同源性相对较低(94.74%)。聚类分析结果也显示ClYVV-JS归入三叶草黄脉病毒分支,与马铃薯Y病毒属其他成员分别归入不同分支。
Clover yellow vein virus(ClYVV),belonging to the genus Potyvirus,infects a variety of legume crops and causes serious damage.In order to clarify the whole genomic sequences and structure characteristics of ClYVV detected in the broad samples collected from Yancheng,Jiangsu province,the genomic fragments were cloned and sequenced.Sequenced fragments were assembled to obtain the full-length sequence of ClYVV-JS.Genomic sequence analysis showed that the genome sequence of isolate was 9585 bp in length,encoding 11 proteins.Sequence alignment results showed that ClYVV-JS shared the highest nucleotide sequence identity(96.37%)with a Japanese ClYVV No.30 isolate(AB011819),and the amino acid sequence identity was 99.06%.While ClYVV-JS shared relative low nucleotide sequence identity(94.74%)with the Chinese Hefei ClYVV isolate(KU922565)reported earlier.The results of cluster analysis also indicated that ClYVV-JS was clustered in the same clade with other ClYVV isolates,while different clade with other members of the genus Potyvirus.
