心安颗粒对慢性心力衰竭大鼠炎性因子、氧化应激、心肌细胞凋亡的影响

The effects of the Xin’an granules on inflammatory factors,oxidative stress and cardiomyocyte apoptosis in rats with chronic heart failure

摘要 目的:通过建立慢性心力衰竭(Chronic Heart Failure,CHF)大鼠模型,探讨心安颗粒对CHF大鼠炎症因子(肿瘤坏死因子-α、白细胞介素-6),氧化应激反应指标(超氧化物歧化酶、丙二醛)、心肌细胞凋亡的影响。方法:SD大鼠60只,随机选50只(余10只为空白对照组),采用腹主动脉缩窄法制备CHF大鼠模型;将存活造模成功大鼠随机分为4组:模型组、贝那普利组、心安颗粒组、贝那普利+心安颗粒组,每组10只;空白对照组不做任何处理,模型组生理盐水灌胃,另外三组分别贝那普利、心安颗粒、贝那普利+心安颗粒灌胃,均每日1次;28 d后测定血清肿瘤坏死因子-α、白细胞介素-6、丙二醛、超氧化物岐化酶含量及心肌细胞凋亡情况。结果:模型组、贝那普利组、心安颗粒组、贝那普利+心安颗粒组与空白对照组比较,肿瘤坏死因子-α、白细胞介素-6、丙二醛明显升高,超氧化物岐化酶明显降低,心肌细胞凋亡增多,差异有统计学意义(P<0.05);贝那普利组与心安颗粒组比较,差异无统计学意义(P>0.05);贝那普利组、心安颗粒组与贝那普利+心安颗粒组比较,差异有统计学意义(P<0.05)。结论:贝那普利联合心安颗粒能显著拮抗CHF大鼠炎症、氧化应激反应,减少心肌细胞凋亡,心安颗粒可与西药相媲美且无明显不良反应,联合用药效果更佳。 Objective:To investigate the effects of Xin’an granules(心安颗粒)on inflammatory factors(TNF-α,IL-6),oxidative stress response(SOD,MDA)and apoptosis of cardiomyocytes in chronic heart failure(CHF)of the rats by establishing the model of chronic heart failure rats.Methods:There were 60 SD rats,50 rats were randomly picked out,and 10 rats of the remaining were blank control group.CHF rat model of the 50 rats was prepared by abdominal aortic coarctation.The rats of successful models were randomly divided into 4 groups:model group,Benazepril group,Xin’an granules group,and Benazepril+Xin’an granules group,with 10 rats in each group.The rats of the blank control group did not do any treatment,the rats of the model group was administered by gavage with normal saline,and the rats of the other three groups were administered by gavage with Benazepril,Xin’an granules,and Benazepril+Xin’an granules by gavage once a day.After 28 days,the levels of TNF-α,IL-6,MDA and SOD in serum were determined and the apoptosis of cardiomyocytes were measured.Results:TNF-α,IL-6 and MDA were significantly increased,SOD was significantly decreased,and apoptosis of cardiomyocytes was increased in the model group,Benazepril group,Xin’an granules group,Benazepril+Xin’an granules group compared with in the blank control group,it were statistically significant(P<0.05).There was no significant difference between Benazepril group and Xin’an granules group(P>0.05).There were statistical significance between Benazepril group,Xin’an granules group and Benazepril+Xin’an granules group(P<0.05).Conclusion:The Benazepril combined with Xin’an granules can significantly antagonize inflammation and oxidative stress,and reduce apoptosis of cardiomyocytes in CHF rats.Xin’an granules can be comparable with western medicine without obvious toxic side effects,and Xin’an granules combined with western medicine is more effective.
