摘要
构建人脐动脉平滑肌细胞(HUASMC)氧化应激模型,采用栝楼籽蛋白酶解物(Protease Hydrolysates of Trichosanthes Seed,PHTS)处理细胞,检测细胞存活率和总谷胱甘肽(GSH)、内皮素(ET)水平,分析过氧化氢酶(CAT)、超氧化物歧化酶(SOD)的基因与蛋白表达。结果表明,栝楼籽蛋白酶解物可有效提升HUASMC细胞的存活率和GSH、CAT、SOD表达水平,抑制ET分泌。
An oxidative stress model of human umbilical artery smooth muscle cells HUASMC is established,and Protease Hydrolysates of Trichosanthes Seed(PHTS)are used to process cells for testing the indexes such as cell survival rate,total glutathione(GSH),endothelin(ET)levels and analyzing the gene expression,protein expression of catalase(CAT)and superoxide dismutase(SOD).The results indicate that PHTS can effectively prolong survival rate of HUASMC cells,expression level of GSH,CAT and SOD,and inhibit ET secretion.
