摘要
目的:研究海风藤醇提物抑制CD31因子对慢性硬膜下血肿(CSDH)模型大鼠血肿外膜血管炎症的影响。方法:将25只大鼠随机分为5组,分别为Sham假手术组,模型组,海风藤醇提物高、中、低剂量组,每组5只。造模结束后连续灌胃21 d,观察各组大鼠的一般行为学、病理组织染色以及大鼠血肿MRI影像。结果:经海风藤醇提物高、中、低剂量灌胃治疗21 d后,各组大鼠偏瘫与癫痫发作均有减少,其中高剂量组体质量增加最为明显(P<0.01),且不良事件发生最少;与模型组、假手术组相比,海风藤醇提物高、中、低剂量组均可见锥体细胞较多且分布规整、紧凑,血肿外膜切片CD31高表达,高剂量组海风藤醇提物中CD31表达明显降低(P<0.05);经大鼠颅脑MRI检测,海风藤醇提物高、中、低剂量组血肿液体积均可见不同程度减少,其中海风藤醇提物高剂量组血肿体积减小最明显(P<0.05)。结论:海风藤醇提物可以抑制CD31炎性介质,从而拮抗CSDH血肿外膜新生血管的生成与崩解,对CSDH起到治疗作用。
Objective:To study the effect of alcohol extract of Caulis Piperis Kadsurae inhibitor CD31 on inflammation of hematoma in CSDH model rats.Methods:25 rats were randomly divided into the sham operation group,the model group,the Caulis Piperis Kadsurae groups of high-dose,medium-dose and low-dose,with 5 rats in each group.After 21 days of continuous gavage after modeling,the general behavior,pathological tissue staining and MRI images of hematoma were observed in different groups.Results:After 21 days of gavage of alcohol extract of Caulis Piperis Kadsurae,the hemiplegia and epilepsy were reduced in each group,among which the weight gain was the most obvious(P<0.05),and the incidence of adverse events was the least in the high-dose group.Compared with the model group and the sham operation group,there were more pyramidal cells with regular and compact distribution in the alcohol extract of Caulis Piperis Kadsurae groups of high-dose,medium-dose and low-dose.The outer membrane biopsy of CD31 was highly expressed under hematoma,and the expression of CD31 decreased significantly in the high-dose group(P<0.05).MRI images showed that the hematoma volume decreased in different degrees in the high-dose,medium-dose and low-dose groups,and the decrease was most significant in the high-dose group(P<0.05).Conclusion:Alcohol extract of Caulis Piperis Kadsurae can inhibit CD31 inflammatory mediators,antagonize the formation and disintegration of adventitia neovascularization in CSDH hematoma,and play a therapeutic role in CSDH.
