急性髓系白血病中SLC39A1表达及其临床意义

Expression of SLC39A1 in acute myeloid leukemia and its clinical significance

目的通过分析SLC39A1基因在急性髓系白血病(AML)中的表达及其临床意义,揭示SLC39A1促进AML发生发展的作用。方法应用GEO数据库(GSE30029)分析90例AML患者和31例健康对照者骨髓细胞SLC39A1基因的表达水平;应用TCGA数据库中172例含随访信息的AML患者病历资料,分析SLC39A1基因表达对预后的影响。结果健康对照组中SLC39A1的中位表达水平为455.1,AML患者中SLC39A1的中位表达水平为585.5,SLC39A1在AML患者中表达上调,差异有统计学意义(P<0.001)。ROC曲线评价结果显示,当截断值为560.7时,诊断AML的敏感度和特异度分别为60.0%和90.3%。SLC39A1高表达组年龄和白细胞数量高于低表达组,差异均有统计学意义(P<0.05);SLC39A1高表达组FAB分型中M4和M5组比率高于低表达组,差异有统计学意义(P<0.001)。SLC39A1高表达组患者的总生存期显著短于SLC39A1低表达组患者,差异有统计学意义(P<0.05)。结论SLC39A1高表达是AML患者中常见的分子事件,SLC39A1高表达与AML患者预后不良相关,并可用于AML疾病进展的监测。

Objective To analyze the level of SLC39A1 expression in acute myeloid leukemia(AML)and its clinical significance,and to reveal the role of SLC39A1 in promoting the development of AML.Methods GEO database(GSE30029)was used to analyze the level of SLC39A1 expression in bone marrow cells of 90 AML patients and 31 healthy control.The data of 172 AML cases with follow-up information in TCGA(The Cancer Genome Atlas)database were applied to analyze the influence of SLC39A1 gene expression on prognosis.Results The median level of SLC39A1 expression in healthy control group was 455.1,while that in AML was 585.5.The expression of SLC39A1 was up-regulated in AML patients,and the difference was statistically significant(P<0.001).The evaluation results of ROC curve showed that when the cutoff value was 560.7,the sensitivity and specificity of AML diagnosis were 60.0%and 90.3%.The age and WBC count of SLC39A1 high expression group were significantly higher than those of SLC39A1 low expression group,and the differences were statistically significant(P<0.01).The percentages of M4 and M5 in the SLC39A1 high-expression group were higher than those in the low-expression group,and the differences were statistically significant(P<0.001).The overall survival time of patients with high expression of SLC39A1 was significantly shorter than that of patients with low SLC39A1 expression,the difference was statistically significant(P<0.05).Conclusion The high expression of SLC39A1 is a common molecular event in AML patients.The high SLC39A1 expression is related to the poor prognosis of AML patients and can be used to monitor the progress of AML diseases.
