摘要
目的:探究Ⅰ型胶原α1链(collagen typeⅠalpha 1,COLIA1)、Ⅰ型胶原α2链(collagen typeⅠalpha 2,COLIA2)在结直肠癌中的表达及其临床意义。方法:抽取南通市第二人民医院普外科2017年9月—2019年3月收治的60例结直肠癌患者作为研究对象,利用实时荧光定量聚合酶链式反应(quantitative reverse transcription polymerase chain reaction,qRT-PCR)技术检测手术切除的结直肠癌组织和癌旁组织中COLIA1、COLIA2 mRNA水平,利用Western Blot技术检测组织中COLIA1、COLIA2的蛋白水平,随访结直肠癌患者预后情况,分析COLIA1、COLIA2在结直肠癌中的表达及其临床意义。结果:结直肠癌患者癌组织中COLIA1、COLIA2 mRNA相对水平显著高于癌旁组织(2.51±0.31 vs 1.00±0.28;4.96±0.51 vs 1.00±0.40),且其蛋白相对含量也显著高于癌旁组织(1.25±0.07 vs 1.00±0.08;1.96±0.26 vs 1.00±0.19)。癌组织中COLIA1、COLIA2的表达与肿瘤直径、临床分期和术后随访结果呈正相关。结论:COLIA1、COLIA2在结直肠癌患者的癌组织中高表达,且与患者预后相关。
Objective:To explore the expression and clinical significance of collagen typeⅠalpha 1(COLIA1)and collagen typeⅠalpha 1(COLIA2)in patients with colorectal cancer.Methods:60 patients with colorectal cancer admitted from September 2017 to March 2019 in the Department of General Surgery were randomly selected as the research objects,and quantitative reverse transcription polymerase chain reaction(qRT-PCR)technology was used to detect surgically resected colorectal cancer tissues and adjacent cancers.The protein levels of COLIA1 and COLIA2 in the tissues were detected by Western Blot technology.Follow up the prognosis of patients with colorectal cancer,and the expression and clinical significance of COLIA1 and COLIA2 in patients withcolorectal cancer were analyzed.Results:The relative levels of COLIA1 and COLIA2 mRNA in cancer tissues of patients with colorectal cancer were significantly higher than those in adjacent tissues(2.51±0.31 vs 1.00±0.28;4.96±0.51 vs 1.00±0.40),and the relative protein level was also significant higher than adjacent tissues(1.25±0.07 vs 1.00±0.08;1.96±0.26 vs 1.00±0.19).The expression of COLIA1 and COLIA2 in cancer tissue was positively correlated with tumor diameter,clinical stage and postoperative follow-up results.Conclusion:COLIA1 and COLIA2 are highly expressed in cancer tissues in patients with colorectal cancer,and are negatively correlated with the prognosis of patients.
