红景天苷通过HIF-1α/NF-κB信号通路对阻塞性睡眠呼吸暂停低通气综合征大鼠炎症的调控作用 被引量:3

摘要 目的研究红景天苷通过低氧诱导因子-1α(HIF-1α)/核因子-κB(NF-κB)信号通路对阻塞性睡眠呼吸暂停低通气综合征(OSAHS)模型大鼠炎症的调控作用。方法36只雄性OSAHS模型大鼠,随机分为正常对照组、慢性间歇低氧组、红景天苷低剂量组、红景天苷高剂量组各9只。正常对照组和慢性间歇低氧组、红景天苷低剂量组、红景天苷高剂量组分别给予生理盐水、低剂量红景天苷、高剂量红景天苷腹腔注射1周,采用酶联免疫吸附试验(ELISA)检测各组大鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-8(IL-8)水平;采用蛋白免疫印迹法(Western blot)检测各组大鼠肺组织HIF-1α及NF-κB蛋白表达,比较各组间细胞因子水平及蛋白表达差异。结果与正常对照组比较,慢性间歇低氧组TNF-α、IL-6、IL-8水平及HIF-1α、NF-κB蛋白表达均升高,差异有统计学意义(P<0.05);应用红景天苷后TNF-α、IL-6、IL-8水平及HIF-1α、NF-κB蛋白表达均下降,红景天苷高剂量组下降更明显,差异均有统计学意义(P<0.05)。结论OSAHS大鼠机体炎症源于慢性间歇低氧,红景天苷可通过HIF-1α/NF-κB信号通路对OSAHS大鼠炎症进行调控,减轻OSAHS大鼠机体炎症。 Objective To study the regulation of salidroside on inflammation through the hypoxia-inducible factor-1α(HIF-1α)/nuclear factor-κB(NF-κB)signaling pathway in obstructive sleep apnea-hypopnea syndrome(OSAHS)model rat.Methods Thirty-six male OSAHS model rats were randomly divided into normal control group,chronic intermittent hypoxia group,low-dose salidroside group,and high-dose salidroside group,9 rats in each group.Normal control group and chronic intermittent hypoxia group,low-dose salidroside group and high-dose salidroside group were given normal saline,low-dose salidroside and high-dose salidroside by intraperitoneal injection for 1week.ELISA was used to examine serum tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and interleukin-8(IL-8)of rats in each group.Western blot was used to examine the protein expression of HIF-1αand NF-κB in lung tissues of rats in each group,and the differences in cytokine level and protein expression among groups were compared.Results Compared with the normal control group,TNF-α,IL-6,IL-8 levels and HIF-1α,NF-κB protein expression increased significantly in chronic intermittent hypoxia group(P<0.05).After salidroside application,TNF-α,IL-6,IL-8 levels and HIF-1α,NF-κB protein expression decreased,especially in high-dose salidroside group,and the differences were statistically significant(P<0.05).Conclusion Salidroside can regulate and reduce the inflammation in OSAHS rats through the HIF-1α/NF-κB signaling pathway.
