摘要
目的探究线粒体钙单向转运体调节因子1(MCUR1)对卵巢癌细胞株HO8910、SKOV3生物学行为的影响。方法将人卵巢癌细胞株HO8910、SKOV3分为四组,一组为空细胞组,其余三组分别转染siRNA干扰序列,检测四组的MCUR1表达水平。选取MCUR1表达水平最低的一组和最高的一组进行后续实验。分析干扰MCUR1对细胞增殖能力、细胞凋亡能力及相关蛋白表达的影响。结果两种细胞株中,MCUR1均在siRNA-2组中表达水平最低。HO8910-siRNA组、SKOV3-siRNA组细胞增殖能力与对应NC组无明显差异(P>0.05)。HO8910-siRNA组的细胞凋亡比例为19.99%,高于HO8910-NC组的7.55%,差异具有统计学意义(P<0.05);SKOV3-siRNA组的细胞凋亡比例为21.49%,高于SKOV3-NC组的5.77%,差异具有统计学意义(P<0.05)。蛋白质免疫印迹(WB)实验结果显示,HO8910-siRNA组、SKOV3-siRNA组的P53表达水平高于对应NC组,差异具有统计学意义(P<0.0001)。结论MCUR1可能通过调控P53通路调控细胞凋亡,有望成为卵巢癌诊断和治疗的新靶点。
Objective To investigate the effects of mitochondrial calcium uniporter regulator 1(MCUR1)on the biological behavior of human ovarian carcinoma cell lines HO8910 and SKOV3.Methods Human ovarian carcinoma cell lines HO8910 and SKOV3 were divided into four groups,one group was empty cell group,the other three groups were transfected with siRNA interference sequence,and the expression level of MCUR1 in the four groups was detected.The group with the lowest expression level of MCUR1 and the group with the highest expression level were selected for subsequent experiments.The effects of interference MCUR1 on cell proliferation ability,apoptosis ability and related protein expression were analyzed.Results In both cell lines,the expression level of MCUR1 was the lowest in the siRNA-2group.There were no significant differences in cell proliferation ability between HO8910-siRNA group,SKOV3-siRNA group and corresponding NC group(P>0.05).The percentage of apoptosis in the HO8910-siRNA group was 19.99%,which was higher than 7.55% in the HO8910-NC group,and the difference was statistically significant(P<0.05);the percentage of apoptosis in SKOV3-siRNA group was 21.49%,which was higher than 5.77% in the SKOV3-NC group,and the difference was statistically significant(P<0.05).Western-blotting(WB)experiment results showed that the expression level of P53 in the HO8910-siRNA group and the SKOV3-siRNA group was higher than that in the corresponding NC group,and the difference was statistically significant(P<0.0001).Conclusion MCUR1 may regulate apoptosis by regulating P53pathway,which may become a new target for diagnosis and treatment of ovarian carcinoma.
