补中益气汤含药血清调控GSK-3β介导的Wnt/β-catenin信号通路增强A549细胞对顺铂敏感性的研究 被引量:4

摘要 目的:探讨补中益气汤含药血清增强A549细胞对顺铂敏感性的机制是否与GSK-3β介导的Wnt/β-catenin信号通路有关。方法:制备补中益气汤含药血清作用于A549细胞株,随机分为a组(对照血清组)、b组(顺铂组)、c组(补中益气汤组)、d组(补中益气汤含药血清联合顺铂组)。流式细胞术检测细胞凋亡,Western blot检测GSK-3β、p-GSK-3β^(ser9)、p-GSK-3β^(tyr216)、β-catenin、survivin蛋白表达。结果:补中益气汤含药血清联合顺铂可降低p-GSK-3β^(ser9)、β-catenin、survivin蛋白表达,提高晚期细胞凋亡率。结论:抑制GSK-3β介导的Wnt/β-catenin信号通路激活可能是补中益气汤含药血清增强A549细胞对顺铂敏感性的作用机制之一。 Objective:To explore whether mechanism of Buzhong Yiqi Decoction containing serum enhancing sensitivity of A549 cells to cisplatin is related to Wnt/β-catenin signaling pathway mediated by GSK-3β.Methods:Buzhong Yiqi Decoction containing serum was prepared and applied to A549 cell line,and randomly divided into:group a(control serum group),group b(cisplatin group),group c(Buzhong Yiqi Decoction group)and group d(Buzhong Yiqi Decoction containing serum combined with cisplatin group).Cell apoptosis was detected by flow cytometry,and expressions of GSK-3β,p-GSK-3β^(ser9),p-GSK-3β^(tyr216),β-catenin and sur⁃vivin were detected by Western blot.Results:Combination of Buzhong Yiqi Decoction containing serum and cisplatin could decrease protein expressions of p-GSK-3β^(ser9),β-catenin and survivin,and increase late apoptosis rate.Conclusion:Inhibiting activation of Wnt/β-catenin signaling pathway mediated by GSK-3βmay be one of mechanisms of Buzhong Yiqi Decoction containing serum enhancing sensitivity of A549 cells to cisplatin.
