摘要
分析比较不同姜黄素产品的功能成分含量和抗氧化活性,为姜黄素产品的品质评价和加工利用提供参考。利用超高效液相色谱法(ultra high performance liquid chromatography,UPLC)测定5种市售姜黄素和实验室自制姜黄素(extracted curcumin,EC)的功能成分姜黄素、脱甲氧基姜黄素和双脱甲氧基姜黄素含量,进一步测定其DPPH、ABTS、超氧阴离子(O_(2)^(-)·)自由基清除能力及总抗氧化能力、脂质过氧化(lipid peroxidation,LPO)抑制活性,最后分析各功能成分含量与抗氧化活性的相关性。结果表明:5种市售姜黄素中总姜黄素含量为1.278~2.188 g·hg^(-1),EC含量则为16.655 g·hg^(-1)。市售姜黄素对DPPH自由基、ABTS自由基、LPO半数清除/抑制浓度(median inhibition concentration,IC_(50))分别为749.68~1568.84μg·mL^(-1)、458.92~851.24μg·mL^(-1)和3.436~6.334 mg·mL^(-1),其对O_(2)^(-)·清除能力为0~2.956 U·g^(-1),总抗氧化能力为0.082~0.156 mmol·g^(-1);EC对DPPH自由基、ABTS自由基、LPO的IC_(50)分别为99.39μg·mL^(-1)、35.49μg·mL^(-1)和0.296 mg·mL^(-1),对O_(2)^(-)·自由基清除能力为20.441 U·g^(-1),总抗氧化能力为1.473 mmol·g^(-1)。姜黄素类化合物含量与抗氧化活性呈极显著正相关关系(P<0.01,0.976≤r≤0.999)。5种市售姜黄素产品的姜黄素类化合物含量和抗氧化活性均存在差异,且显著低于EC,姜黄素类化合物是姜黄素产品抗氧化活性的主要贡献物质。
In order to provide reference for the quality evaluation and utilization of curcumin products,the functional components contents and antioxidant activities of different curcumin products were analyzed and compared.The ultra high performance liquid chromatography(UPLC)was utilized to determine the contents of the functional components such as curcumin,demethoxycurcumin and bisdemethoxycurcumin of 5 kinds of commercial curcumin and curcumin extracted(EC)in the lab.Then,the free radical scavenging ability towards DPPH,ABTS,superoxide anion(O_(2)^(-)·),total antioxidant capacity and the inhibitory activity of lipid peroxidation(LPO)were further determined.Finally,the correlation between the content of each functional component and antioxidant activity was analyzed.The results showed that:the total curcumin content in 5 kinds of commercial curcumin was 1.278-2.188 g·hg^(-1),while the content of EC was 16.655 g·hg^(-1).The median inhibition concentration(IC_(50))of the commercial curcumin towards DPPH free radical,ABTS free radical and LPO were 749.68-1568.84 mg·mL^(-1),458.9_(2)^(-)851.24 mg·mL^(-1)and 3.436-6.334 mg·mL^(-1),respectively,and the scavenging ability towards O_(2)^(-)·was 0-2.956 U·g^(-1),and the total antioxidant capacity was 0.08_(2)^(-)0.156 mmol·g^(-1).By contrast,IC_(50)of EC towards DPPH free radical,ABTS free radical and LPO were 99.39μg·mL^(-1),35.49μg·mL^(-1)and 0.296 mg·mL^(-1),respectively,and the scavenging ability towards O_(2)^(-)·was 20.441 U·g^(-1),and the total antioxidant capacity was 1.473 mmol·g^(-1).There was a significant positive correlation between the content of curcumin derivatives and antioxidant activities(P<0.01,0.976≤r≤0.999).The contents of curcumin derivatives and antioxidant activities in the 5 kinds of commercial curcumin products were different from each other,and lower than EC significantly.And the curcumin derivatives were the main contributing constituents for the antioxidant activities of curcumin products.
