摘要
本试验通过水提醇沉法制备黄芪多糖,并以黄芪多糖和硫酸锌进行制备黄芪锌多糖,并对其体内、体外抗氧化活性进行研究,以期为黄芪锌多糖的开发利用提供理论依据。结果表明,制备的黄芪锌多糖中锌元素含量为(11.32±0.48)mg/g。体外研究结果显示,黄芪多糖和黄芪锌多糖对·OH自由基的清除率分别为43.33%和50.46%;对O2-·自由基的清除率分别为37.91%和48.83%;对1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基的清除率分别为56.86%和72.33%。此外,小鼠体内研究结果表明,黄芪多糖和黄芪锌多糖能显著提高衰老小鼠血清和肝脏中超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)和总抗氧化能力(Total antioxidant capability,T-AOC),降低丙二醛(malondialdehyde,MDA)和过氧化脂质(Lipid peroxide,LPO)水平;且黄芪锌多糖比黄芪多糖具有更强的抗氧化活性。上述结果表明,黄芪锌多糖在体内、体外都表现出良好的抗氧化活性。
In this experiment,astragalus polysaccharide was prepared by water extraction and alcohol precipitation,and astragalus zinc polysaccharide was prepared with astragalus polysaccharide and zinc sulfate,and its in vivo and in vitro antioxidant activities were studied in order to provide theoretical basis for the development and utilization of astragalus zinc polysaccharide.The results showed that the zinc content in the prepared astragalus zinc polysaccharide was(11.32±0.48)mg/g.The results of in vitro studies showed that the scavenging rates of Astragalus polysaccharides and Astragalus zinc polysaccharides to OH radicals were 43.33%and 50.46%,respectively;the scavenging rates of O2-radicals were 37.91%and 48.83%;The scavenging rates of phenyl-2-trinitrophenylhydrazyl(1,1-diphenyl-2-picrylhydrazyl,DPPH)radicals were 56.86%and 72.33%,respectively.In addition,the results of in vivo studies in mice showed that astragalus polysaccharides and astragalus zinc polysaccharides could significantly increase superoxide dismutase(SOD),catalase(CAT)and total antioxidant capacity in serum and liver of aging mice(Total antioxidant capability,T-AOC),reduce malondialdehyde(MDA)and lipid peroxide(Lipid peroxide,LPO)levels;and Astragalus zinc polysaccharide has stronger antioxidant activity than Astragalus polysaccharide.The above results indicated that astragalus zinc polysaccharide exhibited good antioxidant activity both in vivo and in vitro.
