摘要 程序性细胞死亡(programmed cell death,PCD)是由细胞内特定基因调控的分子程序介导的细胞死亡过程,在多细胞生物的正常发育过程和体内稳态维持中发挥至关重要的作用。PARK7基因,编码DJ-1蛋白,最初是作为与RAS基因一起显示转化活性的致癌基因发现的,后来被鉴定为常染色体隐性早发性帕金森病的致病基因,在调节PCD方面有显著作用。DJ-1蛋白可通过正负两种方式调节自噬,通过一系列信号转导途径调节凋亡,还可调节焦亡、铁死亡等过程,在神经退行性疾病、癌症和其他疾病中发挥重要作用。本文探讨DJ-1蛋白在PCD中的作用及分子机制,可为相关疾病的治疗提供新思路。 Programmed cell death (PCD) is a process of cell death mediated by a molecular program regulated by specific genes in the cell.It plays a vital role in the normal development of multicellular organism and the maintenance of homeostasis.The PARK7 gene,encoding the DJ-1 protein,was originally discovered as an oncogene showing transforming activity together with the RAS gene,and was later identified as a causative gene for autosomal recessive early-onset Parkinson’s disease,playing a significant role in regulating PCD.DJ-1 can regulate autophagy in both positive and negative ways,and regulate apoptosis through a series of signal transduction pathways,pyroptosis and ferroptosis,playing an important role in neurodegenerative diseases,cancer and other diseases.The role and molecular mechanism of DJ-1 in PCD were discussed in this paper to provide new ideas for the treatment of related diseases.
