摘要
目的研究美丽薄子木内生真菌Rhytidhysteron sp.BZM-9乙酸乙酯部位的化学成分。方法采用硅胶柱、ODS、Sephadex LH-20凝胶柱色谱和Pre-HPLC进行分离纯化,利用HR-ESI-MS、NMR等方法鉴定化合物的结构。并运用MTT法及96孔板二倍稀释法对部分化合物进行活性评价。结果从菌株Rhytidhysteron sp.BZM-9中分离出15个化合物,分别鉴定为alternariol(1)、porric acid E(2)、3-ethyl-5,7-dihydroxy-3,6-dimethylphthalide(3)、5-羟基-7-甲氧基-4,6-二甲基-2-苯并呋喃酮(4)、6-formyl-7-hydroxy-5-methoxy-4-methylphalide(5)、4-hydroxy-6-methoxy-5-methylphthalide(6)、2-甲基-5-甲氧羰甲基-7-羟基色原酮(7)、2-(2’-hydroxypropyl)-5-methyl-7-hydroxychromone(8)、altechromones A(9)、4-O-methylscleron(10)、pilobolusate(11)、aspergillusphenols A(12)、苔色酸甲酯(13)、1H-indole-3-carboxaldehyde(14)、ergosta-4,6,8(14),22-tetraen-3-one(15)。活性测试结果显示,化合物12对肝癌细胞SMMC7721具有增殖抑制作用(IC_(50)=0.338μmol·L^(-1)),同时对耐甲氧金黄色葡萄球菌也表现出较强的抑制活性(MIC=6.25μg·mL^(-1))。结论所有化合物均为首次从该属真菌中分离得到。化合物12表现出了一定的抗肿瘤、抗菌活性。
Objective To determine the chemical constituents of the ethyl acetate extract of endomycete fungus Rhytidhysteron sp.BZM-9 from Leptospermum brachyandrum.Methods Silica gel,ODS,Sephadex LH-20 column chromatography and Pre-HPLC were used for the isolation and purification.The structures were elucidated on the basis of HR-ESI-MS and NMR spectroscopic evidence.The cytotoxicity and antibacterial activity of some compounds were evaluated by MTT method and double dilution method in the 96-well plate.Results Totally 15 compounds were isolated and identified as alternariol (1),porric acid E (2),3-ethyl-5,7-dihydroxy-3,6-dimethylphthalide (3),5-hydroxy-7-methoxy-4,6-dimethylphthalide (4),6-formyl-7-hydroxy-5-methoxy-4-methylphalide (5),4-hydroxy-6-methoxy-5-methylphthalide (6),2-methyl-5-methoxycarbonylmethyl-7-hydroxychromanone (7),2-(2’-hydroxypropyl)-5-methyl-7-hydroxychromone (8),altechromones A (9),4-O-methylscleron (10),pilobolusate (11),aspergillusphenols A (12),methyl orsellinate (13),1H-indole-3-carboxaldehyde (14),and ergosta-4,6,8 (14),22-tetraen-3-one (15).The activity test showed that compound 12 not only had good proliferation inhibition against liver cancer cells SMMC7721 (IC_(50)=0.338 μmol·L^(-1)),but also significant inhibition against MRSA (MIC = 6.25 μg·mL^(-1)).Conclusion All the compounds are isolated from the genus of Rhytidhysteron for the first time.Compound 12 shows certain antitumor and antibacterial activities.
