Role of cGAS-STING signaling pathway in autoimmune diseases

天然免疫是机体抵御病原体侵害的第一道防线,而机体感知病原体存在则有赖于核酸识别机制。cGAS-STING信号通路是天然免疫系统中重要的模式识别及效应通路之一,主要负责识别存在于胞浆中的DNA分子,并激活下游信号通路产生Ⅰ型干扰素和其他炎症因子。近年来,越来越多的研究表明核酸受体cGAS介导的天然免疫通路cGAS-STING信号通路异常激活与自身免疫性疾病的发生与发展有紧密的联系。本文就cGAS-STING信号通路的激活过程及其在自身免疫性疾病中的相关作用与治疗药物展开综述,旨在为自身免疫性疾病的预防与治疗提供参考。

Innate immunity is the first line of defense against pathogens,and the perception of pathogens depends on the mechanism of nucleic acid recognition.cGAS-STING signaling pathway is an important pattern recognition and effect pathway in the innate immune system,which is mainly responsible for recognizing DNA molecules in the cytoplasm and activating downstream signaling pathways to produce type I interferons and other inflammatory factors.In recent years,more and more studies have shown that abnormal activation of cGAS-STING signaling pathway mediated by nucleic acid receptor cGAS is closely related to the occurrence and development of autoimmune diseases.This article reviewed the activation process of cGAS-STING signaling pathway and its role in autoimmune diseases and therapeutic drugs,to provide theoretical basis and research direction for the prevention and treatment of autoimmune diseases.
