基于双波长指纹图谱和含量测定的经典名方百合地黄汤物质基准的质量评价研究 被引量:1

摘要 目的 建立百合地黄汤物质基准的高效液相色谱法(HPLC)双波长指纹图谱,并测定3种化学成分的含量。方法 采用Kromasil 100-5-C_(18)色谱柱(4.6 mm×250 mm,5 μm);以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱;流速:0.7 mL·min^(-1);柱温:35℃;检测波长:203、320 nm;进样量:10 μL。选用“中药色谱指纹图谱相似度评价系统(2012版)”对15批百合地黄汤物质基准的HPLC色谱图进行分析,建立双波长指纹图谱;同时测定其中3种主要成分的含量。结果 建立了百合地黄汤物质基准的HPLC双波长指纹图谱,波长分别为203 nm和320 nm,其中共有峰分别有16个和9个;并测定了3种化学成分的含量,其中梓醇、地黄苷D和益母草苷的含量分别在36.2126~48.8442 mg·g^(-1)、2.9734~4.8516 mg·g^(-1)和3.6128~5.0232 mg·g^(-1)。结论 本研究通过双波长指纹图谱和含量测定表征了百合地黄汤中鲜百合和鲜地黄汁的信息,为百合地黄汤物质基准及其制剂的质量评价体系建立提供了参考。 Objective To determine 3 chemical components in Baihe Dihuang decoction by HPLC dual-wavelength fingerprint method.Methods Kromasil 100-5-C_(18) column (4.6 mm×250 mm,5 μm) was used.Acetonitrile (A)-0.1% phosphoric acid aqueous solution (B) was used as mobile phase in gradient elution,the flow rate was 0.7 mL·min^(-1),the column temperature was 35℃,the detection wavelength was 203 nm,320 nm,and the injection volume was 10 μL.The “Similarity Evaluation System of Chromatographic Fingerprint of Traditional Chinese Medicine (2012 edition)” was used to analyze the HPLC chromatograms of 15 batches of Baihe Dihuang decoction material reference to establish dual-wavelength fingerprint.The contents of 3 main components were determined simultaneously.Results HPLC dual wavelength fingerprint of Baihe Dihuang decoction was established with the wavelength of 203 nm and 320 nm,and there were 16 and 9 common peaks,respectively.The contents of catalpol,rehmannioside D and leonurin ranged from 36.2126~48.8442 mg·g^(-1),2.9734~4.8516 mg·g^(-1) and 3.6128~5.0232 mg·g^(-1),respectively.Conclusion This study characterized the information of fresh lily bulb and fresh rehmannia root juice in Baihe Dihuang decoction by dual wavelength fingerprint and content determination,providing a reference for the establishment of material reference and preparation quality evaluation system of Baihe Dihuang decoction.
