液相色谱质谱联用测定中毒样品中河豚毒素

Determination of Tetrodotoxin in Poisoned Samples by Liquid Chromatography-Mass Spectrometry

建立血液、尿液、胃液以及鱼样中河豚毒素的液相色谱质谱联用测定方法。鱼样基质样品采用1%甲酸甲醇溶液去蛋白,上清液采用固相萃取法去除杂质,血液、胃液用乙腈提取,尿液静置沉淀,并均采用液相色谱质谱联用仪进行河豚毒素测定。实验结果表明,在线性范围内河豚毒素含量和峰面积具有良好的线性关系。本文所提出的测定中毒样品中河豚毒素的方法具有快速、高效、准确等优点,能够为河豚毒素中毒临床诊断、食品安全检测提供技术保障。

A method for the determination of tetrodotoxin in blood,urine,gastric juice and fish samples by liquid chromatography-mass spectrometry was developed.The matrix of fish sample was deproteinized by 1%formic acid methanol solution,the supernatant was extracted by solid phase extraction,the blood and gastric juice were extracted by acetonitrile,and the urine was deposited,tetrodotoxin was determined by liquid chromatography-mass spectrometry(LC-MS).The results showed that tetrodotoxin content and peak area had a good linear relationship in the linear range.The method is rapid,efficient and accurate,which can be used for clinical diagnosis and food safety detection of tetrodotoxin poisoning.
