摘要
目的探讨C1QA在原发性肝细胞癌(HCC)中的表达及其与临床病理特征的相关性。方法采用免疫组织化学(IHC)染色法检测2018年1月-2022年6月广西医科大学附属肿瘤医院65例原发性肝细胞癌组织及50例癌旁肝组织中C1QA蛋白的表达情况,分析C1QA蛋白的表达与临床病理特征的关系;采用Spearman方法通过GEPIA在线工具中相关性项目分析C1QA与血管内皮标记物及肿瘤相关巨噬细胞的关系。结果C1QA蛋白在原发性肝细胞癌的表达高于癌旁肝组织,但差异无统计学意义(P>0.05);C1QA高表达与原发肝细胞癌微血管弥漫性分布及Edmondson-Steiner分级密切相关(P<0.05),与性别、年龄、民族、肿物长径、脉管癌栓、肝被膜侵犯、近癌旁肝内微血管侵犯、卫星灶情况、肝外转移及HBV感染无关(P>0.05);C1QA与血管内皮标记物CD34及肿瘤相关巨噬细胞(CCR2、CD40及CCL2)的表达水平呈正相关(P<0.05)。结论C1QA基因可能通过促进血管生成及与肿瘤相关巨噬细胞相互作用共同促进原发性肝细胞癌的发展过程,并可能为肝癌的靶向治疗提供新思路。
Objective To investigate the expression of C1QA in primary hepatocellular carcinoma(HCC)and its correlation with clinicopathological features.Methods Immunohistochemical(IHC)staining was used to detect the expression of C1QA protein in 65 cases of primary hepatocellular carcinoma tissues and 50 cases of adjacent liver tissues in the Affiliated Tumor Hospital of Guangxi Medical University from January 2018 to June 2022.The relationship between the expression of C1QA protein and clinicopathological features was analyzed.Spearman method was used to analyze the relationship between C1QA and vascular endothelial markers and tumor-associated macrophages by correlation items in GEPIA online tool.Results The expression of C1QA protein in primary hepatocellular carcinoma was higher than that in adjacent liver tissues,but the difference was not statistically significant(P>0.05).The high expression of C1QA was closely related to the diffuse distribution of microvessels and Edmondson-Steiner grade in primary hepatocellular carcinoma(P<0.05),however,there was no significant correlation with sex,age,nationality,tumor diameter,vascular tumor thrombus,hepatic capsule invasion,intrahepatic microvascular invasion near the cancer,satellite lesions,extrahepatic metastasis and HBV infection(P>0.05).In addition,C1QA was positively correlated with the expression of vascular endothelial marker CD34 and tumor-associated macrophages(CCR2,CD40 and CCL2)(P<0.05).Conclusion C1QA gene may promote the development of primary hepatocellular carcinoma by promoting angiogenesis and interacting with tumor associated macrophages,and may provide a new idea for targeted therapy of hepatocellular carcinoma.
