吡仑帕奈在4~12岁局灶性癫痫患儿中单药治疗的效果及安全性研究

Therapeutic efficacy and safety of perampanel monotherapy in children aged 4-12 years with focal epilepsy

目的探讨吡仑帕奈在4~12岁局灶性癫痫患儿中单药治疗的有效性及安全性。方法收集河南科技大学第一附属医院儿科门诊及郑州大学第一附属医院儿科门诊自2021年8月至2022年1月治疗的8例4~12岁的局灶性癫痫患儿资料,观察吡仑帕奈单药治疗第3个月、第6个月时癫痫发作情况及治疗过程中的不良反应,全部患儿随访6个月。结果共纳入8例患儿,1例患儿治疗10余天时因出现跌倒停药(不计入疗效),1例患儿治疗3个月时因无效自行停药。治疗3个月时有效率为86%(6/7),无发作率为43%(3/7),治疗6个月时有效率为86%(6/7),无发作率为71%(5/7)。1例患儿加药过程中出现困倦、头晕,随着用药时间延长症状自行消失,总不良反应发生率为25%(2/8),随访结束时药物保留率为75%(6/8)。结论吡仑帕奈单药治疗4~12岁局灶性癫痫患儿疗效及耐受性良好。

Objective To explore therapeutic efficacy and safety of perampanel monotherapy in children with focal epilepsy aged between 4-12 years.Methods Clinical data of 8 children aged 4-12 years with focal epilepsy who were treated in the department of pediatrics,the First Affiliated Hospital of Henan University of Science and Technology and the First Affiliated Hospital of Zhengzhou University from August 2021 to January 2022 were collected.The epileptic seizures and adverse reactions during the treatment were observed in the 3rd and 6th months of perampanel monotherapy.All children were followed up for 6 months.Results Among 8 patients,one patient stopped the medication due to falling after more than 10 days of treatment(not included in the curative effect).Perampanel was discontinued in one patient due to ineffectiveness after 3 months of treatment.After 3 months of treatment,therapeutic effective rate was 86%(6/7)and seizure-free rate was 43%(3/7).After 6 months of treatment,therapeutic effective rate was 86%(6/7)and seizure-free rate was 71%(5/7).After taking the medication,one child first developed drowsiness and dizziness,which then spontaneously disappeared with the extension of the medication time.The total adverse reaction rate was 25%(2/8),and the drug retention rate was 75%(6/8)at the end of the follow-up.Conclusion Perampanel monotherapy is effective and well-tolerated in treating children aged 4-12 years with focal epilepsy.
