摘要
目的:探讨抗癌防移片对大肠癌肝转移小鼠模型血清肝细胞生长因子(HGF)、血管内皮生长因子(VEGF)表达的影响。方法:采用脾脏注入CT26细胞的方法制备大肠癌小鼠模型,随机分为假手术组、模型组、抗癌防移片高剂量组、抗癌防移片低剂量组。干预4周后,ELISA法检测血清HGF、VEGF-A表达量,观察各组大肠组织及肝脏组织病理切片。结果:给药2、3、4周,与假手术组比较,其余各组小鼠体重均明显下降(P<0.05),其中模型组下降最显著(P<0.05)。与模型组比较,抗癌防移片高剂量组肝脏瘤体重量降低(P<0.05)。与假手术组比较,模型组小鼠血清CA_(199)、CEA水平均明显升高(P<0.05)。与抗癌防移片高剂量组比较,抗癌防移片低剂量组及模型组小鼠血清CA_(199)、CEA水平均明显升高(P<0.05)。与假手术组比较,模型组小鼠大肠组织及肝组织可见肿瘤细胞浸润;与模型组比较,抗癌防移片高剂量组小鼠大肠组织及肝组织未见明显的肿瘤细胞,细胞形态较为规整。结论:抗癌防移片可通过下调血清HGF、VEGF表达抑制大肠癌肝转移。
Objective:To investigate the effect of Kang’ai Fangyi tablet on the expression of serum hepatocyte growth factor(HGF)and vascular endothelial growth factor(VEGF)in colorectal cancer liver metastasis mouse model.Methods:The colorectal cancer mouse model was prepared by injecting CT26 cells into the spleen,and divided into sham operation group,model group,high-dose Kang’ai Fangyi tablet group,and low-dose Kang’ai Fangyi tablet group randomly.After four weeks of intervention,the expressions of HGF and VEGF-A in serum were detected by ELISA,and the histopathological sections of large intestine tissue and liver tissue in each group were observed.Results:After 2,3,and 4 weeks of intervention,the weight of other groups of mice decreased significantly compared with the sham operation group(P<0.05),and the model group showed the most significant decrease(P<0.05).Compared with the model group,the liver tumor weight in the high-dose group of Kang’ai Fangyi tablet decreased(P<0.05).Compared with the sham operation group,the serum levels of CA_(199) and CEA in the model group were significantly increased(P<0.05).Compared with the high-dose Kang’ai Fangyi group,the serum levels of CA_(199) and CEA in the low-dose Kang’ai Fangyi group and model group were significantly increased(P<0.05).Compared with the sham operation group,tumor cell infiltration was seen in the large intestine and liver tissue of the mice in the model group and the low-dose Kang’ai Fangyi tablet group;no obvious tumor cells were found,cell morphology is more regular.Conclusion:Kang’ai Fangyi tablet can inhibit liver metastasis of colorectal cancer by down-regulating the expression of HGF and VEGF in serum.
